摘要:目的研究党参对慢性阻塞性肺疾病(COPD)的治疗作用及对肿瘤坏死因子-α(TNF-α)/核转录因子-κB(NF-κB)信号通路的影响。方法将70只小鼠用完全随机法分为空白组、模型组、地塞米松组(0.5 mg·kg^(-1))、补中益气丸组(3.6 g·kg^(-1))和低、中、高剂量实验组(2.6,5.2和10.4 g·kg^(-1)党参),每组10只。用经鼻滴注COPD造模液的方法复制COPD小鼠模型,造模的同时按20 mL·kg^(-1)的剂量灌胃给予相应的药物,其中空白组与模型组灌胃20 mL·kg^(-1)纯净水。各组每天给药1次,连续28 d。用肺功能测定系统检测小鼠肺功能,用酶联免疫吸附实验法测定TNF-α含量,用蛋白质印迹(Western blot)法检测肺组织NF-κB的表达水平。结果低、高剂量实验组和地塞米松组、补中益气丸组、模型组、空白组的小鼠呼吸频率分别为(138.1±26.6),(132.8±21.6),(126.7±20.4),(134.7±25.1),(171.5±29.9)和(121.2±15.9)bpm,支气管肺泡灌洗液中TNF-α含量分别为(168.83±19.34),(133.20±15.71),(108.61±13.10),(153.04±17.68),(215.91±25.20)和(58.55±7.69)pg·mL^(-1),肺组织中TNF-α含量分别为(230.86±26.66),(176.16±18.82),(156.83±17.76),(221.40±25.64),(275.62±29.41)和(79.83±8.57)pg·mL^(-1),肺组织NF-κB蛋白相对表达水平分别为0.86±0.03,0.54±0.02,0.45±0.02,0.83±0.03,0.95±0.04和0.38±0.01。低、高剂量实验组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论党参对COPD具有治疗作用,下调TNF-α/NF-κB信号通路而缓解气道炎症是其作用机制之一。
Objective To study the therapeutic effects of Codonopsis Radix on chronic obstructive pulmonary disease(COPD)and its effect on Tumor necrosis factor-α/nuclear factor kappa B(TNF-α/NF-κB)signaling pathway.Methods A total of 70 mice were randomly divided into blank group,model group,dexamethasone group(Dex,0.5 mg·kg^(-1)),Buzhong Yiqi pill group(BYP,3.6 g·kg^(-1))and experimental-L,-M,-H groups(2.6,5.2,10.4 g·kg^(-1) Codonopsis Radix)with 10 cases per group.COPD model mice was replicated by nasal drip of COPD establishing liquid.The blank and model groups were given 20 mL·kg^(-1)purified water once a day for 28 days.The lung function of mice was measured by lung function test system.The content of TNF-α was measured by enzyme linked immunosorbent assay.The expression of NF-κB in lung tissue was detected by Western blot.Results The frequencies of aspiration in the experimental-L,experimental-H,DEX,BYP,model and blank groups were(138.1±26.6),(132.8±21.6),(126.7±20.4),(134.7±25.1),(171.5±29.9)and(121.2±15.9)bpm;the levels of TNF-α in bronchoalveolar lavage fluid were(168.83±19.34),(133.20±15.71),(108.61±13.10),(153.04±17.68),(215.91±25.20)and(58.55±7.69)pg·mL^(-1);the levels of TNF-α in lung tissues were(230.86±26.66),(176.16±18.82),(156.83±17.76),(221.40±25.64),(275.62±29.41)and(79.83±8.57)pg·mL^(-1);the relative expression levels of NF-κB in lung tissues were 0.86±0.03,0.54±0.02,0.45±0.02,0.83±0.03,0.95±0.04 and0.38±0.01.There were significant differences between the model group and the experimental-L,experimental-H groups(P<0.05,P<0.01).Conclusion Codonopsis Radix has therapeutic effect on COPD,one of its mechanisms is related to down regulating TNF-α/NF-κB signaling pathway and alleviating airway inflammation.
