摘要:目的研究大黄牡丹汤组方(RPDP)对急性胰腺炎(AP)模型大鼠胰腺腺泡细胞凋亡的影响。方法大鼠分为药物作用12 h假手术(SO)组、模型组和RPDP组,药物作用24 h SO组、模型组和RPDP组。经十二指肠壁浆肌层穿刺距离胆胰管开口0.3 mm处的胆管,加压注射3.5%牛磺胆酸钠0.1 mL/100 g,建立大鼠AP模型。治疗组造模后立即给予RPDP 40 g/kg灌胃1次,SO组、AP模型组给予等量的生理盐水灌胃1次。经RPDP治疗后12 h和24 h,麻醉取血及胰腺组织,检测胰腺组织细胞凋亡率、一氧化氮(NO)含量和一氧化氮合成酶(iNOS)的活性。结果与模型组比较,RPDP组两个时间点的血清淀粉酶活性显著降低(P<0.05),胰腺组织内NO含量明显升高(P<0.05),胰腺细胞凋亡指数显著增加(P<0.05)。结论 RPDP可诱导急性胰腺炎大鼠胰腺腺泡细胞凋亡,其机制应该与增加胰腺组织iNOS活性和NO含量有关。
Objective To investigate the effects of Dahuang Mudan Decoction prescription(RPDP) on the pancreatic gland cell apoptosis in model rats with acute pancreatitis(AP).Methods Rats were divided into sham operation(SO) group,model group,and RPDP group with drug effect within 12 h and 24 h respectively.AP model group was built through pressure injection of 3.5% sodium taurocholate 0.1 mL /100 g at bile duct where muscular biopsy of duodenal wall of the pulp was at the distance of 0.3mm from bile pancreatic duct opening.After molding,the treatment group was immediately given an intragastric administration of 40 g /kg RPDP,and the SO group and AP model group were given the same amount of normal saline for an intragastric administration.After RPDP treatment of 12 h and 24 h,the rats were anesthetized to get the blood and pancreatic tissue and the apoptosis rate,the content of nitrogen monoxidum(NO) and the activity of inducible nitric oxide synthetase(iNOS) of the pancreatic tissue were detected.Results Compared with the AP model group,the activity of serum-amylase at two time points significant degraded(P〈0.05),the content of nitrogen monoxidummin the pancreatic tissue significant upgraded(P〈0.05).Conclusion RPDP can induce the apoptosis of pancreatic acinar cell in AP rats.The mechanism was probably that it can increase the content of nitrogen monoxidum and the activity of inducible nitric oxide synthetase of the pancreatic tissue.
