摘要:肝癌传统治疗效果有限。铁死亡作为铁依赖的脂质过氧化驱动的细胞死亡方式,在肝癌防治中有重要潜力。内质网-线粒体接触点(EMCS)在铁死亡过程中对脂质转运发挥核心调控作用,其表面富集酰基辅酶A合成酶长链家族成员4、三磷酸肌醇受体-葡萄糖调节蛋白75-线粒体外膜蛋白电压依赖性阴离子通道1复合体等功能蛋白,为脂质过氧化提供关键分子基础。本文综述EMCS结构和铁死亡机制及其在肝癌中的调控网络,阐述EMCS介导脂质转运的分子机制与信号通路,为肝癌治疗新策略的开发提供理论依据。
The treatment of liver cancer remains demanding.Ferroptosis is a type of cell death driven by iron-dependent lipid peroxidation and has significant potential for the prevention and treatment of liver cancer.The endoplasmic reticulum-mitochondria contact site(EMCS)plays a core regulatory role in lipid transport during ferroptosis.Its surface is enriched with acyl-CoA synthetase long-chain family member 4,inositol receptor-glucose regulatory protein 75-mitochondrial outer membrane protein voltage-dependent anion channel 1 complex,etc.,providing a molecular basis for lipid peroxidation.This article reviews the structure of EMCS and mechanism of ferroptosis and its regulatory network in liver cancer,and elaborates on the molecular mechanism of lipid transport mediated by EMCS,providing a theoretical basis for the development of new treatment strategies for liver cancer.
