摘要:目的基于高效液相色谱(HPLC)测定大黄指标成分,研究指标化学成分含量与色度值之间的相关性,建立一种简单、全面和准确的质量评估方法。方法建立HPLC法测定大黄指标成分的方法并测定其含量,利用指纹图谱相似度评价软件进行指纹图谱分析,采用色度与HPLC相结合的研究方法探究化学成分含量与色度的相关性。结果建立13种成分的HPLC测定方法并测定20批样品的指标成分含量,指纹图谱相似度在0.567~0.993;L^(*)值在56.01~80.82,a^(*)值在10.8~29.08,b^(*)值在83.83~104.68;其中L^(*)值、b^(*)值与没食子酸、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚呈负相关。a^(*)值与没食子酸、表儿茶素、芦荟大黄素-8-O-葡萄糖苷、大黄酚-8-O-葡萄糖苷、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚呈正相关等。结论所建立的大黄指纹图谱特征性强,可用于其质量控制,指标成分含量与色度的相关性分析结果有利于建立一种简单、全面和准确的方法用于对大黄质量的评估。
Objective To determine the marker components of Radix et Rhizoma Rhei by high-performance liquid chromatography(HPLC),and to investigate the correlation between the contents of these chemical components and chromaticity values,so as to establish a simple,comprehensive,and accurate method for quality evaluation.Methods An HPLC method was established to determine the contents of marker components.Fingerprint analysis was performed using similarity evaluation software.The correlation between chemical contents and chromatic values was investigated by combining chromaticity measurement and HPLC.Results The established HPLC method was applied to determine the contents of 13 marker components in 20 batches of samples.The fingerprint similarities ranged from 0.567 to 0.993.The L*values ranged from 56.01 to 80.82,a*values from 10.8 to 29.08,and b*values from 83.83 to 104.68.Negative correlations were observed between L*and b*values and the contents of gallic acid,aloe-emodin,rhein,emodin,chrysophanol,and physcion.Positive correlations were found between a*value and the contents of gallic acid,epicatechin,aloe-emodin-8-Oglucoside,chrysophanol-8-O-glucoside,aloe-emodin,rhein,emodin,chrysophanol,and physcion.Conclusion The established fingerprint method exhibits strong specificity and can be used for quality control of Radix et Rhizoma Rhei.The correlation analysis between the contents of marker components and chromaticity values facilitates the development of a simple,comprehensive,and accurate approach for quality evaluation of Radix et Rhizoma Rhei.
