摘要:目的:观察三黄凝胶对大鼠复合耳廓痤疮模型组织中转化生长因子β1(Transforming Growth Factor-β1,TGF-β1)、骨形态发生蛋白7(Bone Morphogenetic Protein 7,BMP-7)表达的影响,探究三黄凝胶预防痤疮瘢痕的作用机制。方法:将32只无特定病原体(Specified Pathogen-free,SPF)级Wistar雄性大鼠按随机数字表法取5只为空白组,其余27只为造模组并建立大鼠复合耳廓痤疮模型,21 d后随机抽取2只做苏木精-伊红(Hematoxylin-eosin,HE)染色并进行镜下观察,确认造模成功后分为模型组、对照组、三黄凝胶高、中、低浓度组,每组各5只;对照组给予维A酸乳膏(每日1.48 g成药),三黄凝胶高、中、低浓度组分别给予不同浓度的三黄凝胶(每日1.1 g、0.568 g、0.3 g生药),空白组涂抹空白凝胶基质(每日0.4 mL凝胶基质),每日1次,连续用药28 d,模型组不予任何处理;在造模第7、14、21天和治疗第14、28天观察各组大鼠右侧耳廓皮损变化;在治疗第28天时采用瘢痕边缘示踪法,测量和计算瘢痕面积;于末次给药24 h后采样,用酶联免疫吸附测定法检测血清中TGF-β1、BMP-7的含量。结果:模型组与空白组比较,TGF-β1表达上升,BMP-7表达降低(P <0.05),且BMP-7表达与TGF-β1呈负相关(r=-0.973,P <0.05);三黄凝胶高浓度组及对照组与模型组比较,TGF-β1表达均有不同程度的降低,BMP-7表达上升(P <0.05)。结论:三黄凝胶能有效干预痤疮瘢痕的形成,可能与其抑制TGF-β1的信号转导,刺激蛋白反馈调节信号通路,促进BMP-7抗纤维化活性有关。
Objective:To observe the effects of the Sanhuang gel(三黄凝胶)on the expressions of TGF-β1 and BMP-7 in rat model of compound auricle acne,in order to explore the mechanism of the Sanhuang gel in preventing acne scar.Methods:5 rats of the 32 SPFgrade Wistar male rats were taken out as the blank group according to the random number table method,and the remaining 27 rats were regarded as the modeling group to establish rats model of composite auricle acne.After 21 days,2 rats in the modeling group were randomly selected for HE stain and observing the image under the microscope.After confirming that the rat models were successfully manufactured,those models were divided into the model group,the control group,the Sanhuang gel high-dose group,the Sanhuang gel medium-dose group and the Sanhuang gel low-dose group,5 rats in each group.The control group was given tretinoin cream(1.48 g patent medicine per day),the Sanhuang gel high-dose,medium-dose and low-dose groups were given different concentrations of the Sanhuang gel(1.1 g,0.568 g,0.3 g crude medicine per day),and the blank group was coated with blank gel matrix(0.4 mL gel matrix per day).The medication was administered once a day for continuous 28 days.Results:Compared with the blank group,TGF-β1 expression in the model group was increased,BMP-7 expression in the model group was decreased(P<0.05),and the expression of BMP-7 was negatively correlated with TGF-β1(r=-0.973,P<0.05).Compared with the model group,the expressions of TGF-β1 in the Sanhuang gel high-dose group and the control group were decreased,and the expressions of BMP-7 in the Sanhuang gel high-dose group and the control group were increased(P<0.05).Conclusion:The Sanhuang gel can effectively interfere in the formation of acne scar,which may be related to its inhibition of TGF-β1 signal transduction,stimulation of protein feedback regulation signal pathway,and promotion of anti-fibrosis activity of BMP-7.
