摘要:目的筛选出适于诱导党参愈伤组织、丛生芽的最佳激素浓度配比。方法取二年生带腋芽的党参茎段作外植体,MS为基本培养基,添加不同浓度配比的6-BA和NAA,研究外植体的愈伤组织和芽的诱导率和生长势。结果适宜诱导愈伤组织的培养基为:MS+6-BA 2.0 mg/L+NAA 1.5 mg/L和MS+6-BA1.0 mg/L+NAA0.5 mg/L,适宜丛生芽诱导和继代的培养基为:MS+6-BA 0.5 mg/L+NAA 0.1 mg/L和MS+6-BA 0.1 mg/L+NAA 0.1 mg/L;6-BA对丛生芽的生长势影响显著,一般低浓度的6-BA配合使用低浓度的NAA有利于丛生芽的诱导和生长。结论该试验为党参种苗的工厂化快速繁殖及基因工程与遗传转化奠定了良好的技术基础。
Objective Induction of callus and cluster buds of Codonopsis pilosula(Franch.)Nannf. were studied.Methods Based on MS medium, stems with axillary buds were in oculated in mediums containing NAA and 6-BA two regulators. Results A good medium for inducing callus is MS + 6-BA 2.0 mg/L + NAA 1.5 mg/l and MS+ 6-BA 1.0 mg/L + NAA 0.5 mg/L;MS+ 6-BA 0.5 mg/L + NAA 0.1 mg/L and MS+6-BA 0.1 mg/L + NAA 0.1 mg/L are good for bud induction. Low concentration of 6-BA and NAA can induce a high ratio of callus. The concentration of 6-BA obviously influences the ratio of bud differentiation. Conclusion To provide scientific basis for rapid production and gene engineering.
