摘要:目的:观察当归多糖对X射线辐射SD大鼠肾脏氧化应激损伤防治作用及其相关机制。方法:48只雄性大鼠随机分为空白对照组、模型组、贞芪扶正颗粒组、当归多糖低、中、高剂量组,共6组,每组8只,贞芪扶正颗粒组灌喂2.62g/kg贞芪扶正颗粒溶液,当归多糖低、中、高剂量组分别灌喂63.6 mg/kg、127.1 mg/kg和254.2 mg/kg当归多糖,空白对照组和模型组分别灌胃等体积的生理盐水,连续灌胃7d后,用X射线6Gy分两次照射,照射后继续灌胃3天。检测各组小鼠体重、肾脏指数变化情况;肾脏HE病理切片;Cr、BUN肾功能结果;GSH-Px、LPO、MDA、SOD氧化应激功能指标和Nrf2蛋白表达情况。结果:与空白对照组相比,模型组大鼠一般状况差,体重下降,肾脏指数升高;Cr和BUN表达升高;LPO和MDA水平增高,SOD、GSH-Px水平降低;Nrf2蛋白表达升高。与模型组相比,63.6 mg/kg、127.1 mg/kg和254.2 mg/kg当归多糖各组大鼠一般状况尚可,体重增加,肾脏指数降低;Cr和BUN表达下降;LPO和MDA表达降低,SOD、GSH-Px水平升高;Nrf2蛋白表达降低。结论:当归多糖对X射线辐射SD大鼠所致的肾脏氧化应激损伤具有防护作用,可能和Nrf蛋白表达有关。
Objective: To study the preventive and therapeutic effect of Angelica polysaccharide(AP) against X-ray radiation inducing oxidative stress in SD rat renal and its related mechanism. Methods: Forty-eight male Sprague Dawley rats were randomly divided into the control group,Zhenqifuzhengkeli group,the low-dose of AP group,the middle-dose of AP group and the high-dose of AP group(with 8 animals in each group),which were administered daily with saline,Zhenqifuzhengkeli at the dose of 2. 62 g/kg and AP at doses of 63. 55 mg/kg,127.1 mg/kg and 254. 2 mg/kg,respectively for seven consecutive days. Then SD rats were exposed to 6 Gy X-ray by two times and after that the administration was continued for 3 days. After the last administration,rats were sacrificed. Then the basic living condition,the weight and kidney index were tested. Morphological changes of testicular cells were observed. Cr and BUN were tested. GSH,LPO,MDA and SOD in rat kidney tissues were measured and the Nrf2 protein expression were tested. Results: Compared with the control group,body weights,SOD and GSH-Px of model rats were significantly decreased; while the kidney index,Cr and BUN,LPO and MDA were increased significantly(P〈0. 05). Compared with the model group,body weights,SOD and GSH-Px of rats in AP group at doses of 63. 55 mg/kg,127. 1 mg/kg and254. 2 mg/kg were significantly increased; the kidney index,Cr and BUN,LPO and MDA were significantly decreased(P〈0. 05). Conclusion: X-ray radiation induces renal oxidative stress damage in SD rats. Angelica polysaccharide has renal protective effects on oxidative stress damage,which may be related to the Nrf2 protein expression.
