摘要:目的探讨当归红芪超滤物对辐射致新生大鼠心肌细胞氧化应激和炎性因子的影响。方法用X线照射新生大鼠原代培养的心肌细胞建立辐射损伤细胞模型,实验分为空白组、模型组、低剂量干预组、中剂量干预组和高剂量干预组,干预组分别用使用3 mg/ml、6 mg/ml和9 mg/ml浓度的当归红芪超滤物对辐射损伤细胞模型进行干预。对各组心肌细胞进行细胞活性检测,超氧化物歧化酶(SOD)、丙二醛(MDA)和乳酸脱氢酶(LDH)等氧化应激指标检测及肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β等炎性因子指标检测。结果各干预组心肌细胞生长抑制率均低于模型组,心肌细胞SOD含量均高于模型组,MDA、LDH含量及TNF-α和IL-1β含量均低于模型组,差异有统计学意义(P<0.05),且呈明显剂量依赖性(P<0.05)。结论当归红芪超滤物可有效改善辐射致新生大鼠心肌细胞的氧化应激及炎性因子指标,避免心肌细胞受损,并与剂量存在密切关系。
Objective To explore the effect of ultra-filtration extract from Angelica sinensis and Hedysarum polybotrys on oxidative stress and inflammatory factors in neonatal rat cardiomyocytes induced by radiation.Methods X-ray irradiation was used on the primitively cultured neonatal rat cardiomyocytes to set up radiation damage models.The experiment was divided into blank,model,low-,medium-and high-dose intervention groups.The radiation damage models were respectively intervened with ultra-filtration extract from Angelica sinensis and Hedysarum polybotrys at concentrations of 3,6,and 9 mg/ml.The cardiomyocyte indicators of cell activity,oxidative stress(SOD,MDA and LDH),and inflammatory factors(TNF-αand IL-1β)in all groups were detected.Results The inhibition rate of cardiomyocyte growth in each intervention group was significantly lower than that of model group(P<0.05),the content of SOD was significantly higher than that of model group(P<0.05),and the contents of MDA,LDH and TNF-αand IL-1βwere significantly lower than those of model group(P<0.05).Conclusions The ultra-filtration extract from Angelica sinensis and Hedysarum polybotrys could effectively improve the indicators of oxidative stress and inflammatory factor in neonatal rat cardiomyocytes induced by radiation and avoid myocardial cell damage,which were closely related with the dose of ultra-filtration extract.
