摘要:目的:用反相高效液相色谱法同时测定湿生扁蕾中木犀草素、当药黄素和当药醇苷的含量。方法:色谱柱:Agilent SB-C18(250 mm×4.6 mm,5μm);流动相:在0~20 min内,甲醇∶水(含0.04%H3PO4)由40∶60~63∶37线性梯度洗脱,在20~50 min内,甲醇∶水(含0.04%H3PO4)=63∶37等度洗脱;流速:0.8 m L/min;检测波长:254 nm,柱温:30℃。结果:木犀草素在30 min内达到基线分离,回归方程:Y=84 800X+4540(r=0.999 9),线性范围0.848 0~4.240μg;当药黄素在15 min内达到基线分离,回归方程:Y=52 300X-32 300(r=0.999 9),线性范围0.896 0~4.480μg;当药醇苷在20 min内达到基线分离,回归方程:Y=102 000X+83 400(r=0.999 9),线性范围为0.832 0~4.160μg。结论:该测定方法快速,测定结果准确可靠。
Objective: To simultaneously determine luteolin,swertisin and swertianolin in Gentianopsis paludosa by reverse high performance liquid chromatography. Methods: By Agilent SB- C18( 250 mm × 4. 6 mm,5 μm) chromatographic column,mobile phase: methanol and water( 0. 04% H3PO4),methanol: water( in 0 ~ 20 min 0. 04% H3PO4),linear gradient elution by 40 ∶ 60 ~ 63 ∶ 37 within 20 ~ 50 min; methanol: water( 0. 04% H3PO4) = 63 ∶ 37 isocratic elution;flow rate of 0. 8 m L / min,the detection wavelength of 254 nm,the column temperature 30 ℃. Results: Luteolin baseline separation was achieved within 30 min. The regression equation: Y = 84 800 X + 4 540( r = 0. 999 9),the linear range0. 848 ~ 4. 240 μg. The swertisin baseline separation was achieved within 15 min,the regression equation: Y = 52 300X-32 300( r = 0. 999 9),the linear range 0. 896 ~ 4. 480 μg. The swertianolin baseline separation was achieved within 20 min,the regression equation: Y = 102 000 X + 83 400( r = 0. 999 9) and the linear range was 0. 832 ~ 4. 160 μg. Conclusion: The method is rapid,accurate and reliable.
