摘要:目的探讨西黄丸对乳腺增生大鼠乳腺组织形态和免疫功能的影响。方法 60只大鼠随机均分为正常组、模型组、他莫昔芬组(0.0018g·kg^(-1))和西黄丸低、中、高剂量组(0.54、1.08、2.16g·kg^(-1))。除正常组外,各组大鼠均肌肉注射苯甲酸雌二醇和黄体酮以诱导乳腺增生模型,复制模型同时给药组灌胃相应药物,正常组和模型组灌胃等体积蒸馏水。游标卡尺测量大鼠乳头直径和高度;Elisa试剂盒测定大鼠血清中雌二醇、孕酮和白细胞介素-2、肿瘤坏死因子-α水平,计算脏器指数(胸腺、脾脏),HE染色并观察大鼠乳腺组织形态学变化,采用立体定量分析方法观察乳腺小叶体密度、间质体密度和腺泡比表面。结果西黄丸低、中、高剂量可显著升高乳腺增生大鼠血清中白细胞介素-2、孕酮水平(P<0.05,P<0.01),降低肿瘤坏死因子-α、雌二醇水平(P<0.05,P<0.01);大鼠胸腺、脾脏指数显著增大(P<0.05,P<0.01);乳腺小叶体密度、腺泡比表面降低(P<0.05,P<0.01),小叶间质体密度增加(P<0.05,P<0.01)。结论西黄丸能够抑制大鼠乳腺增生,其作用机制可能与调节激素水平、增强免疫清除功能相关。
Objective To explore the effect of Xihuangpills on mammary tissue morphology and immune function of rats with mammary gland hyperplasia.Methods Sixty rats were randomly divided into normal group,model group,tamoxifen group(0.0018 g·kg^-1)and Xihuangpill low,middle and high dose groups(0.54,1.08,2.16 g·kg^-1).The model of mammary gland hyperplasia was induced by estradiol benzoate and progesterone in each group except the normal group.The replicated models were administered with corresponding drugs,while the normal group and model group were given distilled water.The levels of Estradiol(E2),Progesterone(P)and IL-2,TNF-αin the serum of rats were measured with Elisa kit.The organ indexes(thymus,spleen)and HE staining were calculated,the morphological changes of the mammary gland were observed,and the density of breast lobules,interstitial density and the acinar surface were observed with stereotaxic analysis.Results Xihuangpills low,middle and high dose obviously increased the levels of IL-2 and P(P〈0.05,P〈0.01),decreased the levels of TNF-αand E2(P〈0.05,P〈0.01)in serum of rats with mammary gland hyperplasia,and improved the indexes of the thymus and spleen(P〈0.05,P〈0.01).Pathological morphology showed that the density of breast lobules and the acinar surface of acinar were significantly decreased(P〈0.05,P〈0.01),while the interstitial density was increased distinctly(P〈0.05,P〈0.01).Conclusion Xihuangpills can inhibit mammary gland hyperplasia in rats,its mechanism may be related to the regulation of hormone levels and enhance immune dearance.
