摘要:目的:探究紫花高乌头总生物碱对肺腺癌A549细胞增殖、凋亡、迁移及细胞周期的影响。方法:采用MTT比色法检测紫花高乌头总生物碱对肺腺癌A549细胞增殖的抑制作用;采用倒置显微镜观察细胞的生长形态;采用细胞划痕试验检测细胞的迁移能力;采用流式细胞仪检测细胞的凋亡情况与细胞周期的分布。结果:与空白对照组比较,紫花高乌头总生物碱浓度在5~320 mg/L之间时对A549细胞增殖有显著的抑制作用(P<0.05),最佳作用浓度为80 mg/L,半数抑制浓度(IC_(50))为73.15 mg/L;紫花高乌头总生物碱20、40、80 mg/L浓度组的细胞凋亡率显著上升(P<0.05);紫花高乌头总生物碱40、80 mg/L浓度组的划痕愈合率显著降低(P<0.05);紫花高乌头总生物碱20、40、80 mg/L浓度组G_(0)/G_(1)期的细胞比例显著升高(P<0.05),S、G_(2)/M期的细胞比例显著降低(P<0.05)。与卡铂组(20 mg/L)比较,紫花高乌头总生物碱80 mg/L浓度组对A549细胞增殖抑制、凋亡、迁移及细胞周期的影响差异均无统计学意义(P>0.05)。结论:紫花高乌头总生物碱可抑制体外肺腺癌A549细胞增殖,其机制可能与诱导A549细胞凋亡、抑制A549细胞迁移以及促进细胞G_(0)/G_(1)周期阻滞有关。
Objective:To study the effect of alkaloids from Aconitum septentrionale on cell proliferation,apoptosis,migration and cell cycle of lung adenocarcinoma A549 cells.Methods:The inhibitory effect of alkaloids from A.septentrionale on cell proliferation of lung adenocarcinoma A549 cells was detected by MTT colorimetric assay,and cell morphology was observed by inverted microscope.Wound healing method was used to detect the migration of A549 cells.The cell apoptosis and cell cycle were analyzed by flow cytometry.Results:Compared with the blank control group,when the concentrations of alkaloids from A.septentrionale were between 5 and 320 mg/L,it had a significant inhibitory effect on the proliferation of A549 cells(P<0.05).The optimal concentration was 80 mg/L,and the half maximal inhibitory concentration was 73.15 mg/L.The apoptosis rates of the groups with the concentrations of 20,40 and 80 mg/L of alkaloids from A.septentrionale increased significantly(P<0.05).The scratch healing rates of the groups with the concentrations of 40 and 80 mg/L of alkaloids from A.septentrionale decreased significantly(P<0.05).The proportions of cells in the G_(0)/G_(1)phase in the groups with the concentrations of 20,40 and 80 mg/L of alkaloids from A.septentrionale increased significantly(P<0.05),while the proportions of cells in the S and G_(2)/M phases decreased significantly(P<0.05).Compared with the carboplatin group(20 mg/L),there were no significant differences in the effects of the group with the concentration of 80 mg/L of alkaloids from A.septentrionale on the proliferation inhibition,apoptosis,migration and cell cycle of A549 cells(P>0.05).Conclusion:Alkaloids of A.septentrionale can inhibit the proliferation of lung adenocarcinoma A549 cells in vitro,and its mechanism may be related to inducing the apoptosis of A549 cells,inhibiting the migration of A549 cells and promoting the cell cycle arrest in the G_(0)/G_(1)phase.
