摘要:目的:探讨菊粉通过激活磷酸腺苷激活的蛋白激酶(AMPK)改善代谢综合征大鼠胰岛素抵抗。方法:80只6周龄雄性Wistar大鼠随机分为两组。正常对照组(Nor)喂以普通饲料,代谢综合征组(MS)喂以高脂高糖饲料,于第11周给予MS组一次性腹腔注射链脲佐菌素(STZ,35 mg/kg)诱导代谢综合征大鼠模型。造模成功后,随机将MS组大鼠分为模型对照组、菊粉高剂量组、菊粉低剂量组、盐酸罗格列酮组。通过血清学检测甘油三酯、胆固醇、高密度脂蛋白、低密度脂蛋白、空腹血糖。采用酶联免疫吸附法测定空腹胰岛素(INS)、脂联素(APN),并计算大鼠胰岛素抵抗指数(HOMA-IR)。用蛋白质印迹法(Western Blot)检测肝脏组织中AMPK相关蛋白表达。用HE染色观察大鼠肝脏病理变化。结果:菊粉高剂量组较模型对照组血脂、血糖、空腹胰岛素水平显著降低(P〈0.05),AMPK蛋白表达水平显著升高(P〈0.05)。结论:菊粉能提高代谢综合征大鼠的胰岛素抵抗能力,控制血糖、血脂水平,上调AMPK的蛋白表达。
Objective :To study the effect of inulin ameliorating insulin resistance via AMPK activation inmetabolic syndrome rats. Methods :Eighty male wistar rats aged of 6 weeks divided randomly into nor-mol control group and metabolc syndromn group,the nor group was fed with regular chow,MS group was fed with the high fat diet. At 11 weeks,the rats of MS group were injected by a low dose of STZ(35 kg/kg)once,and the metabolic syndrome rats model were stablished. Then the MS rats were randomly divided into four groups: the control group,inulin high dose group,inulin low dose group,rosiglitazone group. To estimate triglyceride,cholesterin,high density lipoprotein,low density lipoprotein and fasting blood-glucose by the rats serum. To estimate fasting insulin by the enzyme linked immunosorbent assay,and insulin resistance was calculated using homeostasis model assessment of insulin resistance(HOMA-IR)in rats. To estimate expression of AMPK by using Western Blot. And to observe the pathological changes of the rat liver tissue by the HE staining. Results :Compared with the metabolic syndrome group,inulin high dose group had lower lever of lipid metabolism and glycemic,and higher protein lever of AMPK. Conclusion :Inulin can improve insulin resistance,decrease the lever of blood lipids and glycemic,and regulate up the expression of AMPK.
