摘要:目的建立羌活感冒颗粒中葛根素和黄芩苷的含量测定方法。方法采用双波长高效液相色谱(HPLC)法测定羌活感冒颗粒中葛根素和黄芩苷的含量,色谱柱为Thermo Hypersil Gold-C18柱(5μm,4.6 mm×250 mm),流动相为乙腈-0.5%磷酸,梯度洗脱,流速1.0 m L/min,检测波长葛根素为250 nm、黄芩苷为278 nm,柱温30℃。结果葛根素回归方程为Y=3.9×107X+60 453(r=0.999 6),黄芩苷为Y=3.3×107X+68 288(r=0.999 6),表明葛根素在0.014 88~0.148 80 mg/m L、黄芩苷在0.027 36~0.273 60 mg/m L范围内呈良好的线性关系。葛根素平均回收率为97.95%,黄芩苷为96.33%;葛根素RSD为1.0%,黄芩苷RSD为0.9%。结论所建立的方法专属性强,重复性好,稳定易行,可用于同时测定羌活感冒颗粒中葛根素和黄芩苷的含量。
Objective To establish content test method for puerarin and baicalin in Qianghuo Ganmao Particle.Methods The content of puerarin and baicalin in Qianghuo Ganmao Particle was determined by dual-wavelength HPLC. The samples were separated on the column of Thermo Hypersil Gold-C(18)( 5 μm,4.6 mm×250 mm),which moving phase was acetonitrile-phosphoric acid( 0. 5%). The flow rate was 1. 0 m L/min,the detected wavelengths were set at 250 nm and 278 nm respectively and the column temperature was set at 30 ℃. Results The regression equation of puerarin was Y = 3.9×107 X+60 453( r = 0.999 6),showing a good linearity in the range of 0.014 88 to 0.148 80 mg/m L; while the regression equation of baicalin was Y = 3.3×107 X+68 288( r = 0.999 6),showing a good linearity in the range of 0.027 36 to 0.273 60 mg/m L. The recovery rates of puerarin and baicalin were 97.95% and 96.33%; while RSD were 1.0% and 0.9% respectively. Conclusion The method is stable and feasible with strong specificity and repeatability,which can be simultaneously used to determine the content of puerarin and baicalin in Qianghuo Ganmao Particle.
