摘要:目的研究黑逍遥散含药血清对Aβ25-35/H2O2分别诱导的PC12细胞模型酶学指标的影响。方法经体外培养的大鼠肾上腺嗜铬肿瘤细胞(PCl2细胞)分为空白对照组(常规培养液)、Aβ25-35/H2O2损伤组(常规培养液+Aβ25-35或H2O2)、黑逍遥散含药血清组(常规培养液+Aβ25-35或H2O2+不同浓度的黑逍遥散含药血清)。MTT检测细胞各组存活率,比色法检测LDH、T-SOD、MDA酶学指标。结果 Aβ25-35/H2O2损伤组细胞存活率均显著低于空白对照组(P<0.05),黑逍遥散血清组细胞存活率均高于损伤组,以10%含药血清组最为明显(P<0.05);黑逍遥散含药血清可提高LDH、T-SOD活性,降低MDA含量,但以10%组最为明显(P<0.05)。结论在两次实验中,黑逍遥散均能够不同程度的保护PC12细胞,其机制可能和抗氧自由基有关。
Objective:To study the influence of medicated serum of Black Xiaoyao Powder on PC12 cell model enzymology indexes induced by Aβ25-35/H2O2 respectively. Methods :The rat adrenal gland ehromaffin tumor ceils (PC12 cells)cultured in vitro were divided into a blank control group( conventional culture), Aβ25-35/H2O2 injury group( conventional cultures + Aβ25-35 or H2O2 ), Black Xiaoyao Powder medicated serum group( conventional cultures + Aβ25-35 or H2O2 + different Black Xiaoyao Powde medicated serum). MTT was used to determine cell survival rate of each group, eolorimetry used to detect enzymology indexes of LDH, T -SOD and MDA. Results:The cell survival rate of Aβ25-35/H2O2 injury group was significantly lower than that of the blank control group( P 〈 0. 05 ). The cell survival rate of Black Xiaoyao Powder serum group was higher than that of the injury group, the most obvious with 10% medicated serum group (P 〈 0.05). The medicated serum of Black Xiaoyao Powder improved the activities of LDH and T - SOD and reduced the content of MDA, but most obviously with 10% group (P 〈0. 05). Conclusion:In the two experiments, Black Xiaoyao Powder can protect PC12 ceils at different level and its mechanism may be related to anti - oxygen free radical.
