摘要:卵巢癌(ovarian cancer,OC)是女性癌症相关死亡的主要原因之一,>80%的OC患者会产生多药耐药(multidrug resistance,MDR),这导致了OC的不良预后。三磷酸腺苷(adenosine triphosphate,ATP)结合盒(ATP-binding cassette,ABC)家族,即ABC转运蛋白相关的耐药机制是公认的产生MDR的原因之一。各种ABC转运蛋白在化疗后出现异常表达增加,导致化疗药物在细胞内的积聚减少,从而影响化疗效果,减少OC患者生存期。虽然目前对于ABC转运蛋白抑制剂的临床试验尚无明确结论,但ABC转运蛋白抑制剂的研究对于完善OC治疗仍具有一定意义。了解ABC转运蛋白的功能及其在OC化疗耐药中的作用,对开发更有效的靶向药物具有现实意义。本研究针对ABC转运蛋白中ABCA亚群及ABCB亚群中的一些常见成员对OC的影响进行综述。
Ovarian cancer(OC)is one of the leading causes of cancer-related death in women.More than 80%of OC patients develop multipledrug resistance,which leads to a poor prognosis of OC.The adenosine triphosphate(ATP)binding cassette(ABC)family,also called the drug resistance mechanism associated with ABC transporters,is a recognized cause of multidrug resistance.The abnormal expression of various ABC transporters after chemotherapy leads to the decrease of the accumulation of chemotherapeutic drugs in cells,which affects the results of chemotherapy and reduces the survival time of OC patients.Although there is no clear conclusion on the clinical trials of ABC transporter inhibitors at present,the study of ABC transporter inhibitors still has a certain significance for improving the treatment of OC.Understanding the function of ABC transporter and its role in OC chemotherapy resistance is of practical significance for the development of more effective targeted drugs.This article reviews the effects of ABCA subsets of ABC transporters and some common members of ABCB subsets on OC.
