摘要:目的观察秦艽不同配伍药对对风寒湿痹和风湿热痹模型大鼠血清肿瘤坏死因子α(TNF-α)和白细胞介素1(IL-1β)水平的影响,并探讨其治疗痹证的作用机制。方法将128只Wistar大鼠随机分为风寒湿痹和风湿热痹两大组,每组再分为空白组、模型组、雷公藤组、单味秦艽组、秦艽桑寄生组、秦艽防己组、秦艽威灵仙组、秦艽羌活组,每组8只。采用佐剂诱导加外界施以"风寒湿"和"风湿热"致病因素制备风寒湿痹与风湿热痹大鼠模型,并自造模第1天起各给药组给予制备的相应药液10ml/kg灌胃,空白组和模型组给予等量生理盐水,每日1次,连续25天。测定各组大鼠血清TNF-α和IL-1β水平。结果风寒湿痹和风湿热痹模型组大鼠TNF-α、IL-1β水平均高于空白组(P<0.01)。风寒湿痹和风湿热痹中各给药组大鼠血清TNF-α、IL-1β较模型组均有不同程度降低(P<0.05或P<0.01)。风寒湿痹模型中,秦艽羌活组、秦艽威灵仙组、秦艽桑寄生组大鼠的TNF-α、IL-1β水平下降更明显(P<0.01);秦艽羌活组、秦艽威灵仙组、秦艽桑寄生组大鼠的TNF-α水平低于秦艽防己组(P<0.05或P<0.01)。风湿热痹模型中,秦艽防己组、秦艽桑寄生组、秦艽羌活组大鼠的TNF-α、IL-1β水平下降更明显(P<0.01);秦艽防己组大鼠的IL-1β水平低于秦艽威灵仙组(P<0.05)。结论秦艽配伍羌活、威灵仙、桑寄生降低风寒湿痹模型大鼠TNF-α、IL-1β水平效果明显,秦艽配伍防己降低风湿热痹模型大鼠TNF-α、IL-1β水平效果明显。
Objective To observe the effects and mechanism of Qinjiao (Radix Gentianae macrophyllae) in different combi- nations on the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-lβ (IL-1β) in wind-cold-damp or wind-damp- heat arthralgia model rats. Methods Totally 128 Wistar rats were randomized into two major groups, i.e. the wind-cold-damp arthralgia group and wind-damp-heat arthralgia group, and then each group was further divided into the blank group, model group, Tripterygium group, single Qinjiao group, Qinjiao-Sangjisheng (Herba Taxilli) group, Qinjiao-Fangji (Radix Stephani- ae Tetrandrae) group, Qinjiao-Weilingxian (Radix et Rhizoma Clematidis) group and Qinjiao-Qianghuo (Rhizoma et Radix No- topterygii) group, with 8 in each. Wind-cold-damp arthralgia or wind-damp-heat arthralgia models were induced in all groups except for the blank groups by adjuvant and being exposed in wind-cold-heat or wind-damp-heat environment, respectively. From the 1st day of modeling, each administration group was gavaged with 10 ml/kg corresponding drug solution and the blank and model groups were given equivalent normal saline once daily for 25 consecutive days. The serum levels of TNF-α and IL-Iβ were detected. Results The serum levels of TNF-α and IL-Iβ in the wind-cold-damp arthralgia group and wind-damp-heat ar- thralgia group were significantly higher than those in the blank group (〈P0. 01). The serum levels of TNF-α and IL-Iβ in all administration groups were significantly decreased in varying degrees (P〈0.05 or P〈0.01), especially in the Qinjiao-Qiang- huo group, Qinjiao-Weilingxian group and Qinjiao-Sangjisheng group of the wind-cold-damp arthralgia models (P〈0.01). The TNF-α levels in the Qinjiao-Qianghuo group, Qinjiao-Weilingxian group and Qinjiao-Sangjisheng group were significantly lower than that in the Qinjiao-Fangji group (P〈0.05 or P〈0.01). The serum levels of TNF-α and IL-Iβ were significantly de- creased in the Qinjiao-Fangji group, Qinjiao-Sangjisheng group and Qinjiao-Qianghuo group of wind-damp-heat arthralgia mod- els (P〈0.01). The IL-β level in the Qinjiao-Fangji group was significantly lower than that in the Qinjiao-Weilingxian group (P〈0.05). Conclusion The combinations of Qinjiao-Qianghuo, Qinjiao-Weilingxian and Qinjiao-Sangjisheng can obviously decrease the serum levels of TNF-α and IL-Iβ in the wind-cold-damp arthralgia model rats and the combination of Qinjiao-Fangji can obviously decrease the serum levels of TNF-α and TNF-β in the wind-damp-heat arthralgia model rats.
