摘要:目的建立一种评价多波长色谱指纹图谱的新方法——均谱法(MFM),用于高乌头药材不同极性部位HPLC指纹图谱研究,为全面评价高乌头药材的质量提供参考。方法采用系统溶剂法对不同产地高乌头药材95%乙醇提取物进行萃取,依次得到石油醚、三氯甲烷、乙酸乙酯、正丁醇、乙醇和水6个不同的溶剂萃取部位;采用HPLC法分别于235,254,280,290,308 nm紫外波长下检测,将每个萃取部位5个波长下HPLC图谱的信号加和后求平均值作为该部位的均谱;采用中药指纹图谱相似度评价系统(2012版)对10批高乌头药材不同溶剂萃取部位HPLC均谱进行共有峰确认及相似度评价。结果建立了高乌头药材不同溶剂萃取部位的多波长指纹图谱(MWCF),高乌头石油醚、三氯甲烷、乙酸乙酯、正丁醇、乙醇及水提部位MWCF分别确定了36,26,34,24,16,19个共有峰,均指认了2个指纹峰(高乌甲素和冉乌头碱)。不同产地高乌头药材同一极性部位MWCF相似度较高,说明10批不同产地高乌头药材基源一致,且提示提取和萃取工艺相对稳定;不同极性部位之间化学成分有明显差异,共有峰数目从多到少,高乌甲素和冉乌头碱从有逐渐变到无,充分表征了有效成分在各个萃取部位的分布特征。结论所建的均谱法能够使高乌头药材化学指纹信息最大化,能够最大限度地表征高乌头药材指纹图谱特征,不同溶剂萃取部位MWCF可以全面反映高乌头药材的化学成分分布,为高乌头药材的整体质量评价提供参考。
Objective To establish a mean fingerprint method(MFM),parallel multiple wavelength chromatographic fingerprints(MWCF),for HPLC fingerprints analysis of different extracts of Aconitum sinomontanum Nakai (ASN),and provide basis for quality control of ASN.Methods The experimental drugs(petroleum ether,chloroform,ethyl acetate,n-butanol,alcohol and water extracts)were obtained by systematic solvent extraction from the 95%ethanol extract of ASN,MWCF of the each extract was established by HPLC at wavelength of 235,254,280,290,308 nm,respectively.Mean value of the signals of the same extract from ASN at five wavelengths were obtained and used as the mean chromatographic fingerprint of the extract.Similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(2012 edition)was used to confirm the common peaks and evaluate the similarity in HPLC mean fingerprints analysis of different extracts of ASN.Results The common profiles for MWCF of different extracts of ASN were established.Thirty-six,26,34,24,16,and 19 common peaks were determined in MWCF of petroleum ether extract,chloroform extract,ethyl acetate extract,n-butanol extract,alcohol extract,water extract,respectively.And 2 peaks(lappacontine and ranaconitine)in all extracts were identified.The high similarity of fingerprint of the same ASN extracts indicated that source of 10 batches of different areas were the same,and the processing of the extraction was relatively stable.The distribution of the active chemical composition of different extracts were fully reflected by significant differences in chemical compositions among different extracts,the total numbers of common peaks from more to less,and the content of lappacontine and ranaconitine from something to nothing.Conclusion MFM can maximally retain the fingerprint peaks,achieve fingerprint information,effectively improve fingerprint signal quality,and fully reflect the distribution of the chemical composition of ASN,provide a reference for the comprehensive quality evaluation of ASN.
