摘要:[目的]观测红芪多糖对糖尿病大鼠早期视网膜中核转录因子-κB影响。[方法]使用随机平行对照方法,60只大鼠喂养1周,称重后随机数字表法分为6组,空白对照组、模型组、羟苯磺酸钙组、红芪多糖高剂量组、红芪多糖中剂量组、红芪多糖低剂量组,10只/组。适应喂养1周后,造模大鼠均喂以高糖高脂饲料,第2周所有动物禁食不禁水12h以上,以剂量(30mg/Kg)链脲佐菌素(streptozotocin,STZ),溶于0.1mol/L柠檬酸缓冲液,p H4.4,制成浓度为1%溶液,左侧下腹部注射,连续注射3d,72h后尾静脉取血测血糖(BG),BG≥16.7mmol/L为糖尿病模型诱导成功;高糖高脂饮食继续喂养。模型组:生理盐水,1次/d;羟苯磺酸钙组:多贝斯,90mg/kg,1次/d;红芪多糖高、中、低剂量组:分别灌胃红芪多糖150mg/kg、100mg/kg及50mg/kg,均1次/d。观测视网膜切片NF-κB免疫组化染色灰密度定量、体重、血糖。[结果]初始血糖、干预后血糖、灰度值模型组、羟苯磺酸钙组、红芪多糖高、中、低剂量组均大于空白对照组(P<0.05,P<0.01);初始血糖灰度值模型组、羟苯磺酸钙组、红芪多糖高、中、低剂量组与模型组无明显差异(P>0.05);干预后血糖羟苯磺酸钙组、红芪多糖低组与模型组无明显差异(P>0.05),红芪多糖低组与羟苯磺酸钙组无明显差异(P>0.05);灰度值红芪多糖高组与羟苯磺酸钙组无明显差异(P>0.05)。光学显微镜下观察发现,NF-κB在正常组主要表达于视网膜神经节细胞、血管内皮细胞的细胞质,表达较弱。在给药5周后,模型组大鼠视网膜上NF-κB的表达较正常组明显增加,细胞核着染成深棕黄色,在内核层和外核层呈微弱阳性表达。红芪多糖高、中、低剂量组与西药组均较模型组明显降低NF-κB蛋白的表达。[结论]红芪多糖可降低糖尿病大鼠早期视网膜NF-κB表达。
[Objective] To observe the effect of Hedysarum Polybotys saccharide(HPS) on NF-κB expression in early retina of experimental diabetic rat. [Methods] 60 rats were randomly divided into 6 groups: normal control group, model control group, calcium dobesilate control group, high-dose HPS group, middose HPS group and low-dose HPS group, 10 rats were randomly divided into groups. Adapt to feed after 1 week, the model control rats duplicated by high-fat and high-sugar foods, were intraperitionedl injected by a streptozotocin30mg/Kg, and dissolved in 0.1mol/Lsodium citrate buffer, PH4.4, made in a concentration of 1%,continuous administration during the Three days. After 72 hours, then the rats whose Fasting blood glucose(FBG) equals or surpasses 16. 7mmol/L were selected and divided into model control group, calcium dobesilate control group, high-dose HPS group, mid-dose HPS group and low-dose HPS group, which were treated respectively by 0.9% Na CI, calcium dobesilate(90mg/kg), high-dose HPS(150mg/kg), mid-dose HPS(100mg/kg) and low-dose HPS(50mg/kg) once daily. To observe Gray scale value of NF-κB immunohistochemical in rat retinal sections, weight, Fasting blood glucose. [Results] The initial blood glucose value, the blood glucose value after intervention, the gray value of the rest of the rats were greater than or less than the normal control group(P〈0.05,P〈0.01); Compared with model control group, The initial blood glucose value of HPS each dosegroup and calcium dobesilate control group had no significant difference(P〈0.05);Compared with model control group, the calcium dobesilate control and the low-dose HPS group after the intervention of blood glucose had no significant difference(P〈0.05);The calcium dobesilate control and the high-dose HPS group of retinal gray scale had no significant difference.(P〈0.05). By using the optical microscope and observations, NF-κB in the normal group was mainly expressed in the cytoplasm of retinal ganglion cells and vascular endothelial cells, and the expression was weak. After 8 weeks of administration, compared with the normal group, the expression of NF-κB in the retina of model group was significantly higher, the nuclei are stained dark brown yellow, the expression of NF-κB was weakly positive in the inner nuclear layer and outer nuclear layer. Compared with the model group, HPS each dose group and calcium dobesilate control group of NF-κB expression reduced more significantly. [Conclusion] Hedysarum Polybotys saccharide can reduce the amount of expression of NF-κB in early retina of diabetic rat.
