摘要:目的:探讨敦煌固本方对运动性疲劳小鼠自由基代谢及DNA损伤的影响。方法:昆明种小鼠60只,随机分为6组(n=10),分别为安静对照组(C)、安静给药组(E)、运动对照组(TC)、运动给药组(TE),运动组又各分为力竭运动后即刻组和力竭运动后恢复24 h组,即TC0、TC24、TE0、TE24。用单细胞凝胶电泳(SCGE)技术检测小鼠外周血细胞DNA损伤,同时测定血清中SOD、GSH-Px活性和MDA含量。结果:①E组小鼠血清中SOD活性明显高于C组(P<0.05),而MDA含量和GSH-Px活性无明显差异(P>0.05);TE0组小鼠血清SOD、GSH-Px活性均显著高于TC0组(P<0.05),而MDA含量明显低于TC0组(P<0.05);TC24和TE24组各指标均趋于安静时水平,GSH-Px、SOD活性和MDA含量均有显著差异(P<0.05)。②各组外周血细胞DNA损伤率,E组与C组相比,无明显差异(P>0.05);TE0与TC0组均高于C组(P<0.05),TE0组明显低于TC0组(P<0.05);TC24组增加达最高值,而TE24组同TE0组相比略有增加,但差异性不显著(P>0.05)。结论:敦煌固本方具有清除自由基、抗氧化的功效,同时对力竭运动导致的小鼠外周血细胞DNA的损伤具有保护作用。
Objective: To evaluate the effect of Dunhuang Guben prescription on blood free radical metabolism and DNA damage in mice induced by exercise fatigue.Methods: 60 Kunming male mice were randomly divided into six groups(10 mice for each group) : rest control group(C),rest experimental group(E),exercise training control group(TC),exercise training experimental group(TE).The trained mice were dived into immediately and 24-hour !s recovery after exhaustive exercise subgroups respectively,such as TC0,TC24,TE0,TE24.We used single cell gel electrophoresis(SCGE) to detect the DNA damage of peripheral blood cell,meanwhile we measured the change of MDA,SOD and GSH-Px of the mice.Results: ①The activities of SOD were higher than those of control group in rest state(P &lt; 0.05),while MDA concentrations and GSH-Px activities were not significant(P &gt; 0.05).In immediately after exhaustive exercise,the blood !s SOD,GSH-Px activities were obviously higher and MDA concentration was remarkable lower in experimental groups than those in control groups(P &lt; 0.05).After 24-hours recovery,SOD,GSH-Px and MDA had a significant difference compared experimental group with its control group(P &lt; 0.05).②In rest state,the DNA damage rate of peripheral blood cell in the experimental group didn’t have a significant difference compared with the control group(P &gt; 0.05).The DNA damage was higher in immediately after exhaustive exercise than that in rest state(P &gt; 0.05).It was obviously lower in TE0 than those in TC0(P &lt;0.05).After 24-hour’s recovery,the DNA damage rate was the highest in TC24.But it was not significant compared with TE0 and TE24(P &gt;0.05).Conclusion: Dunhuang Guben prescription has a strong effect on anti-oxidation and eliminating free radical.Dunhuang Guben prescription has a good effect on protecting blood cell from DNA damage.
