摘要:目的研究逍遥散对氢溴酸东莨菪碱致阿尔茨海默病模型小鼠血清及脑组织匀浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)水平的影响,探讨逍遥散防治阿尔茨海默病的作用及其机制。方法将50只SPF级小鼠随机分为空白组,模型组,逍遥散低、中、高剂量组5组,以腹腔注射氢溴酸东莨菪碱复制阿尔茨海默病小鼠模型,并以不同剂量的逍遥散进行干预防治,给药第10天腹腔注射氢溴酸东莨菪碱1 h后取血及脑组织测血清及脑组织匀浆中SOD,GSH-PX活性及MDA水平。结果模型组小鼠血清及脑组织匀浆中SOD与GSH-PX活性显著降低,MDA水平显著升高,与空白组比较差异有高度统计意义(P<0.01);逍遥散低、中、高剂量均可显著升高小鼠血清及脑组织匀浆中SOD与GSH-PX活性,降低MDA水平,与模型组比较差异有统计意义(P<0.05或P<0.01),但以逍遥散高剂量组作用最为显著(P<0.05)。结论逍遥散对阿尔茨海默病具有较好的防治作用,改善抗氧化能力可能是其作用机制之一。
Objective To study the effects of Xiaoyao San on the activity of SOD and GSH-PX and the level of MDA in serum and brain tissue homogenate of Alzheimer' s disease model mice, and investigate the effect and mechanism of Xiaoyao San treating Alzheimer' s disease. Methods 50 SPF mice were randomly divided into blank control group, model group and low, middle, high dose group of Xiaoyao San. The Alzheimer' s disease mice model was induced with scopolamine hydrobromide alkali intraperitoneal injection, and prevented by different dosage of Xiaoyao San. The activity of SOD and GSH-PX and the level of MDA in serum and brain tissue homogenate were detected after 1 hour injecting scopolamine hydrobromide on the 10th day giving drug. Results Compared with blank control group, the activity of SOD and GSH-PX in serum and brain tissue homogenates of model group were significantly decreased,and the level of MDA was significantly increased(P〈0.01 ). Compared with model group, low,middle,high dose group of Xiaoyao San could significantly increase the activity of SOD and GSH-PX and de- crease the level of MDA(P〈0.05 or P〈0.01 ),but high dose group of Xiaoyao San was most obvious( P〈0.05 ). Conclusion Xiaoyao San has better preventive and therapeutic effect on Alzheimer' s disease, improvement of an- tioxidant capacity may be one of its mechanisms.
