摘要:为了探讨中药增骨丸对去卵巢大鼠血清及骨髓上清中白细胞介素 1β(IL 1β)的影响及血清和骨髓上清中IL 1β的关系。将 4 8只 8月龄Wistar雌性大鼠随机分为正常对照组 ,模型组 ,乙烯雌酚组及三种不同浓度的增骨丸组。每组 8只。正常组仅摘除一段肠系膜 ,其余 5组做卵巢切除术。术后 1周开始给药 ,连续用药 90天后取血和右侧股骨。分离血清及骨髓上清 ,分别测定血清及骨髓IL 1β的含量。结果显示 ,卵巢切除后大鼠血清及骨髓上清中IL 1β的含量均明显增高 ,而经过雌激素及中药治疗能明显降低卵巢切除所致的IL 1β的增高 ,且血清与骨髓上清中IL 1β的含量呈总体正相关 ,但骨髓上清中IL 1β的含量明显高于血清。表明中药增骨丸通过抑制IL 1β而防治骨质疏松 ,这是该药的作用机理之一。血清骨髓上清的变化比较一致 。
The purpose of the paper is to explore the influence of TCD “ZENGGU WAN” (ZGW, a TCD pills for increasing BMD) on interleukin-1β (IL-1β) in blood and bone marrow of ovariectomized rats and to study the relationship between IL-1β in blood and IL-1β in bone marrow. Forty-eight female rats aged 8 months were randomly divided into six groups: the normal, the modeled, the estrogen-treated and three ZGW-treated groups for 3 different concentrations (n=8 rats each). The normal group was given mesenteric excision and the rest 5 groups were given ovariectomy. One week postoperatively, the animals began to be given the medicines for 90 days. Then the blood and the right femora were taken, the serum and the marrow supernate were separated, and the IL-1β levels in the serum and in the marrow supernate were determined, respectively. The results showed that after ovariectomy, the IL-1β levels in rat serum and marrow supernate were obviously increased, but through the treatment by estrogen or by TCD, the IL-1β increase resulting from ovariectomy could be obviously lowered, the IL-1β level in the serum was positively correlated to that in the marrow supernate in the whole, but the latter was obviously higher than the former, suggesting that ZGW could prevent and treat osteoporosis through its inhibiting IL-1β, which is one of the action mechanism of the drug. The IL-1β levels in the blood serum and in the marrow supernate have the same change but the IL-1β level in the marrow supernate has more significance.
