摘要:目的:观察电针结合中药对抑郁模型大鼠HPA轴的影响,探讨中药以及针药结合的抗抑郁作用及其机制。方法:将70只雄性大鼠随机分为正常组、抑郁模型组、西药组、中药组、电针组、电针加中药组、电针加西药组,每组10只。除正常组外,其余均采用慢性不可预见性应激刺激诱导产生大鼠抑郁模型,应激刺激的第8天,治疗组在造模的同时接受治疗,每天1次,共21天。实验第28天,处死动物,取血,采用放射免疫分析法测定大鼠血清皮质醇(CORT)和促肾上腺皮质激素(ACTH)含量。结果:抑郁模型组大鼠血清CORT和ACTH含量较正常组有显著升高(P<0·01);各治疗组血清CORT、ACTH含量均显著下降(P<0·05,0·01),除电针组、中药组外,其余各组的CORT水平与正常组比较无显著性差异,各治疗组比较没有显著差异。结论:慢性应激致抑郁大鼠的HPA轴功能亢进,各治疗方法均可通过抑制亢进的HPA,下调血中ACTH、CORT以发挥抗抑郁作用,针药结合存在着一定的优势。
Objective:To observe the effect of electro-acupuncture combining with medicine on HPA axis of rats with chronic unpredictable stress,approach the mechanism.Methods:70 male rats were randomly divided into seven groups,normal,model,western medicine,Chinese herbs,electro-acupuncture,electro-acupuncture plus Chinese herbs and electro-acupuncture plus western medicine groups.The stress models were induced by chronic unpredictable stimulation.Each group was given the corresponding treated for 21 days after modeling eight days,the serum ACTH and CORT contents were determined by radioimmunoassay.Results:The serum ACTH and CORT contents in model group rats were obviously higher than those in normal group,they were significantly lowered in each treated group,there was no difference between normal and each treated group on the serum ACTH and CORT contents except simple electro-acupuncture and Chinese herbs groups.Conclusion:Electro-acupuncture combining with medicine could effectively treated depression,inhibiting the hyperaction of HPA axis to down-regulate serum ACTH and CORT contents maybe its mechanism.
