摘要:目的:通过I L-2、I L-6的测定,探索白花蛇舌草总黄酮抑制肿瘤的机理及苯并(a)芘对机体免疫功能的影响。方法:将SPF级昆明种小鼠60只随机分为对照组20只及苯并(a)芘实验组40只,再将苯并(a)芘实验组随机分为苯并(a)芘10 mg/kg组、苯并(a)芘20 mg/kg组,每组20只,雌雄各半分笼饲养,并以二甲亚砜溶解苯并(a)芘,然后分别以苯并(a)芘10 mg/kg和20 mg/kg,腹腔注射,1次/d,连续注射10次,以DMSO100μL/只腹腔注射作为对照,2个月后将苯并(a)芘10 mg/kg组、苯并(a)芘20 mg/kg组再分为白花蛇舌草总黄酮高、低剂量组2组(乙醇回流法提取白花蛇舌草总黄酮),每组10只,分别给予白花蛇舌草流浸膏20 g/kg、10 g/kg灌胃,每天一次,连续给药2周后取材。采用ELI SA法测定血清中I L-2及I L-6;取肝、脾、肾、胰、胃肠、肺等器官,常规包埋,切片,HE染色,显微镜下观察。结果:I L-2浓度,对照组、苯并(a)芘10 mg/kg组、苯并(a)芘20 mg/kg组分别为(360±15.92)ng/L、(92.59±6.12)ng/L、(83.84±5.89)ng/L,实验组明显低于对照组;蛇草总黄酮高剂量组高于低剂量组。I L-6浓度,对照组为(32.65±5.32)pg/m L,苯并(a)芘10mg/kg组、20 mg/kg组分别为(21.78±4.62)pg/ml、(24.54±9.89)pg/m L,与对照组比较差异无显著性;蛇草总黄酮高、低剂量组比较差异无显著性。苯并(a)芘对肝脏、胃、肾脏有明显损害,癌前病变率与对照组比较均有显著差异;对胃的损害,苯并(a)芘20 mg/kg组明显高于苯并(a)芘10 mg/kg组,呈明显的量效关系;实验组肺脏、胰腺均未见明显损害。结论:实验各组I L-2明显低于对照组,提示苯并(a)芘对机体的细胞免疫有明显抑制作用;苯并(a)芘对肝脏、胃和肾脏均有明显损害;胃癌发生率呈明显的量效关系。白花蛇舌草总黄酮高剂量组I L-2浓度明显高于低剂量组,而I L-6浓度无显著差异,提示白花蛇舌草总黄酮有一定的细胞免疫调节功能,可能通过调节T细胞功能,对抗化疗药物的骨髓抑制而实现。
Objective: To explore the mechanism of total flavonoids extracted from BaiHua SheSheCao (Hedyotis diffusa Willd) inhibiting tumor and the influence of benzo (a)pyrene on the immunity of the body by detecting the levels of IL-2 and IL-6. Methods:Sixty mice were randomly divided to 20 ones in the control group and 40 in the experiment group of benzo (a)pyrene, 40 mice in the experiment group of benzo (a)pyrene were randomized into two groups:10 mg/kg benzo(a)pyrene group and 20 mg/kg benzo(a)pyrene group, with 20 mice in each group, they were kept in the cage with half male and half female, and they accepted peritoneal injection of ben-zo (a)pyrene in the concentrations of 10mg/kg and 20mg/kg after dissolving benzo (a)pyrene in dimethyl sulphoxide (DMSO), once per day, for ten consecutive times, in the control group, each rat received peritoneal injection of DM-SO, 100 mL, The experiment groups of benzo(a)pyrene with the concentrations of 10mg/kg and 20mg/kg were divid-ed into high dosage group and low dosage groups of total flavonoids extracted from BaiHua SheSheCao in two months (total flavonoids extracted from BaiHua SheSheCao by ethanol refluxing method), ten mice each group, and they were perfused with BaiHua SheSheCao liquid extract in the concentrations of 20 g/kg and 10 g/kg, once per day, the tissues were obtained after two consecutive weeks. ELISA method was to detect the levels of IL-2 and IL-6 in the serum; the organs including liver, spleen, kidney, pancreas, stomach, lung and others were taken out and fixed in formaldehyde solution, embedded, sectioned, stained and observed under microscope. Results:The concentrations of IL-2 in the control group, 10mg/kg benzo(a)pyrene group and 20mg/kg benzo(a)pyrene group were respectively (360.00± 15.92) ng/L, (92.59±6.12) ng/L and (83.84±5.89) ng/L, the experiment groups were lower than the control group sig-nificantly; high dosage group of total flavonoids extracted from BaiHua SheSheCao was lower than that of low dosage group. The concentrations of IL-6 in the control group, 10mg/kg benzo (a)pyrene group and 20mg/kg benzo (a)pyrene group were respectively (32.65±5.32) pg/mL, (21.78±4.62) pg/mL and (24.54±9.89) pg/mL, the differ-ences was insignificant when they were compared with the control group;the difference was insignificant between high dosage group and low dosage group of total flavonoids extracted from BaiHua SheSheCao. Benzo (a)pyrene could harm liver, stomach and kidney evidently, compared with the control group, the difference was significant in precancerous lesion rate;20mg/kg benzo(a)pyrene group was higher than 10mg/kg benzo(a)pyrene group clearly in stomach harm, it showed dose-effect relationship significantly;there was no obvious harm to lung and pancreas in the experiment group. Conclusion:Different experiment groups are lower than the control group in the concentra-tions of IL-2, which indicates that benzo (a)pyrene could inhibit cellular immunity of the body notably; benzo (a) pyrene could harm liver, stomach and kidney obviously;the incidence of gastric cancer indicates dose-effect relation-ship clearly. High dosage group of total flavonoids extracted from BaiHua SheSheCao is higher than low dosage group in the concentrations of IL-2, while the comparison had no difference in the concentrations of IL-6, it suggests that total flavonoids extracted from BaiHua SheSheCao could regulate cellular immunity to a certain extent, the mechanism might be related to regulating the functions of T cells and fighting against bone marrow arrest of chemotherapeutics.
