摘要:目的:观察黄芩水煎液对CCl4致肝损伤小鼠肝线粒体的影响,揭示黄芩保肝的作用机制。方法:将48只小鼠,随机分为正常对照组、模型组、联苯双酯对照组、黄芩组。正常对照组、模型组每日给予0.2 m L/10 g生理盐水,联苯双酯组每日给予0.150 mg/g联苯双酯,黄芩组每日灌服6 g/kg黄芩水煎液,连续灌胃7天。末次给药2小时后,除正常对照组外,其余各组均腹腔注射0.1%CCl4橄榄油溶液0.1m L/10 g建立小鼠肝损伤模型,正常对照组腹腔注射等体积橄榄油。18小时后,剖取肝脏,检测肝线粒体呼吸链复合体Ⅰ、Ⅳ酶活性,观察肝线粒体超微结构损伤。结果:黄芩水煎液可明显增强小鼠肝线粒体呼吸链复合体Ⅰ、Ⅳ酶活性(P<0.05);电镜观察显示,模型组线粒体肿胀,嵴缩短、消失,基质出现空泡变性;黄芩组多数线粒体超微结构正常。结论:黄芩水煎液可通过增强线粒体呼吸链复合体Ⅰ、Ⅳ酶活力及稳定和保护线粒体的结构,改善CCl4致小鼠肝脏能量代谢障碍,而起到保护肝脏的作用。
Objective: To study the effects of Scutellaria baicalensis Georgi decoction on hepatic mitochondrion in liver injury mice,and explore the mechanism of protection of Scutellaria baicalensis Georgi on liver injury. Methods: 48 mice of SPF grade were randomly divided into four groups: normal control group,acute liver injury model group,positive control bifendatatum group,Scutellaria baicalensis Georgi group. Mice in normal control group and model group were given saline( 0. 2 m L /10g) by intragastric administration; bifendatatum group mice were given bifendatatum( 0. 150 mg / g); Scutellaria baicalensis Georgi group mice were given Scutellaria baicalensis Georgi decoction( 6 g / kg) lasting for seven days. Two hours after the last medication,mice in all groups were injected 0. 1% CCl4 olive oil( 0. 1 m L /10 g) by intraperitoneal to induce the acute liver injury model except the normal control group administrated with same volume normal saline. Eighteen hours later,the liver tissue of each group were collected to detected the activities of complexⅠand Ⅳ of hepatic mitochondria respiratory chain by spectrophotometer,and the hepatic mitochondria ultrastructure were observed with electron microscope.Results: the activities of complexⅠand Ⅳ of hepatic mitochondria respiratory chain were both elevated obviously after the mice were given Scutellaria baicalensis Georgi decoction( P〈0. 05). The ultrastructure observation showed mitochondrion swelling,ciste decrease,repture and disappearance,vacuolar degeneration in the model group. Most of mitochondrion ultrastructure were normal in Scutellaria baicalensis Georgi group. Conclusion:Scutellaria baicalensis Georgi decoction can enhance the activities of complexⅠand Ⅳ of hepatic mitochondria respiratory chain,protect mitochondrion ultrastructure,and improve energy metabolite dysfunction,Scutellaria baicalensis Georgi decoction have protective effect on liver.
