摘要:目的探讨化浊颗粒对非酒精性脂肪肝病(NAFLD)大鼠模型血脂代谢的干预作用。方法将50只大鼠随机分为空白组、模型组、罗格列酮组、化浊颗粒高、低剂量组各10只,空白组以标准饲料喂养;其余大鼠以高脂饲料喂养8周建立NAFLD大鼠模型,罗格列酮组给予罗格列酮片0.36mg/kg灌胃,化浊颗粒高、低剂量组分别给予化浊颗粒2.025、0.675g/kg灌胃,模型组予等量生理盐水灌胃,均每日1次。治疗4周后观察各组大鼠体重、肝指数、总胆固醇(TC)、甘油三酯(TG)、肿瘤坏死因子(TNF-α)、肝细胞脂肪变性的变化。结果化浊颗粒高剂量组大鼠治疗后体重较治疗前降低(P<0.01);与模型组相比,化浊颗粒高剂量组及罗格列酮组肝指数、TG、TC、TNF-α均降低(P<0.05或P<0.01)。结论化浊颗粒可降低NAFLD大鼠TG、TC、TNF-α,从而改善NAFLD大鼠的血脂代谢异常和病理改变。
Objective To discuss the intervention effect of Huazhuo Granules (Granules for dissolving turbidity) on the lipid metabolism of rat models with non-alcoholic fatty liver disease (NAFI.D). Methods Fifty rats were randomized into blank group, model group, rosiglitazone group, and Huazhuo Granules high-dose and low-dose group. The blank group was fed with standard feed, and the rest were given high fat feed for 8 weeks for setting up NAFLD models. Rosiglitazone group was given rosiglitazone 0.36mg/kg for gastric perfusion. Huazhuo Granules high-dose and low-dose group were given the granules 2. 025 and 0. 675g/kg respectively for gastric perfusion. The model group was given normal saline of the same volume. All groups were observed on the changes of weight, liver index, total cholesterol (TC), triglyceride (TG), free fatty acid (FFA), tumor necrosis factor α (TNF-α), and fatty degeneration of hepatocytes. Results The body weight of Huazhuo Granule hige-dose group decrease significantly after treatment (P〈 0.01). Compared with the model group, the Huazhuo Granules high-dose group and the rosiglitazone group showed decrease in liver index, TG, TC and TNF-α (P〈0.05 or P〈0.01). Conclusion Huazhuo Granules is effective in lowering TG, TC, and TNF-α of NAFLD rat models, thus improved their abnormal lipid metabolism and pathological changes.
