摘要:目的:研究超滤技术应用于大黄总蒽醌的纯化工艺。方法:用水提取大黄,提取液离心分离得滤液,以超滤压力、药液温度、料液浓度为考察因素,以总蒽醌含量为考察指标,采用均匀设计优化超滤工艺参数。结果:优化得到大黄总蒽醌超滤工艺为药液以3 000 r/min离心处理12分钟,用分子截留量100 K的膜,超滤压力为0.05~0.09MPa,药液温度25~40℃,药液浓缩程度1∶10~1∶30,pH6.0,超滤膜先用tween-80(5g/L)处理10分钟。结论:优化所得超滤工艺与未超滤相比有较高的总蒽醌得率和纯度。
Objective: To study the purification of total anthraquinone in Chinese Rhubarb with ultrafiltration. Method: Ultrafiltration process was optimized with homogeneous design by adopting total anthraquinone as index, pressure, temperature and the concentration as parameters. Chinese rhubarb was extracted with water and the extract was obtained with centrifuge. Result: The optimal condition of purification process for total anthraquinone in Chi- nese rhubarb was: the liquid centrifuged for 12minutes at the speed 3 000r/min, with ultrafiltration membrane of 100 K retention molecular weight, the pressure 0.05 to 0.09 Mpa and the temperature 25 to 40℃, the concentration ratio 1 : 10 to 1 : 30, pH 6.0, the ultrafiltration membrane was handled with tween-80 (5 g/L) for 10 minutes. Conclusion: Compared with non-ultrafiltration, the yield and pureness of total anthraquinone obtained from the optimized ultrafil- tration orocess are hi^er.
