摘要：目的:构建一种符合中医证候特点且与人溃疡性结肠炎(Ulcerative Colitis,UC)相似的脾虚湿困型UC大鼠模型。方法:采用病证结合的方法,在建立脾虚湿困型Wistar大鼠的基础上,用2,4-二硝基苯磺酸(2,4-Dinitrobenzenesulfonic acid,DNBS)+乙醇溶液灌肠,分别于灌肠后7d、14d、21d取大鼠结肠。通过大鼠证候学、结肠黏膜损伤及镜下观察的改变,评价病症结合动物模型的制作。结果:脾虚湿困型UC组大鼠在造模第3天开始出现饮食减少,倦怠懒动,蜷缩扎堆,毛色枯槁易脱落,且便质变软,甚至有黏液,DNBS+乙醇灌肠后有脓血便,体质量下降明显。镜下观察见结肠黏膜组织结构破坏,有大量炎性细胞浸润,累及肌层,伴有局部肉芽组织增生的表现。结论:病证结合DNBS+乙醇诱导的脾虚湿困型UC大鼠模型类似于人的UC,且符合中医证候特点。
Objective:A model of ulcerative colitis rats of Pixu Shikun type which is similar to human ulcerative colitis was built.Methods:On the basis of the establishment of Wistar rats with the Pixu Shikun type,the rats were enema with2,4-dinitrobenzenesulfonic acid and ethanol,and the colon was taken at7d,14d and21d after enema.Through the rat syndrome,colonic mucosal injury and the change by microscopic observation,the association of the disease with the animal model was evaluated.Results:In the third day,reducing the diet,burnout lazy,huddle up,dull fur,soft stool,and even mucus were fund in UC group of the Pixu Shikun type.Bloody stools appeared when the rats were enema with DNBS and ethanol,and weight was decreased significantly.By microscopic observation,colonic mucosal tissue structure was damaged;a large number of inflammatory cell infiltration were found,involving the muscularis,accompanied by local granulation tissue hyperplasia performance.Conclusion:The UC models with the Pixu Shikun type induced by DNBS and ethanol are similar to human UC and accords with TCM syndrome.
