摘要:目的:观察黄芪甲苷对特发性肺纤维化(IPF)模型大鼠肺组织碱性成纤维细胞生长因子(b FGF)表达的影响,为其临床应用开发提供依据。方法:气管穿刺快速推注博来霉素法复制大鼠肺纤维化模型。将SPF级Wistar大鼠按随机数字表法分为空白组,模型组,激素组,黄芪甲苷高、中、低剂量组,每组12只。造模后第2日起,各组给予不同干预,分别于灌胃后第7、14、28天,采用免疫组化结合图像分析处理系统,比较b FGF在大鼠肺组织中的表达。结果:模型组肺组织b FGF的表达在7、14、28天较同期空白组增强(P<0.05);黄芪甲苷组、激素组表达较同期模型组明显减弱(P<0.05);黄芪甲苷高、中剂量组肺组织b FGF的表达较激素组减弱(P<0.05);低剂量组表达与激素组比较差异无统计学意义(P>0.05)。结论:黄芪甲苷可以抑制特发性肺纤维化模型大鼠肺组织b FGF的表达,其干预效果与剂量呈正相关。
Objective: To provide the evidence for clinical application and development of astragaloside Ⅳ by observing its influence on basic fibroblast growth factor(b FGF) in lung tissue of the rats with idiopathic pulmonary fibrosis(IPF). Methods: Rat model was replicated by tracheal puncturing and quick pumping bleomycin. According to random number table method, SPF Wistar rats were allocated to the blank group, the model group, the hormone group, high, moderate and low doses groups of astragaloside Ⅳ, 12 rats each group. All the groups accepted different intervention since the second day after successfully establishing the models. The expressions of b FGF in the lung tissue of the rats were compared by adopting immunohistochemical methods combined with imaging analysis processing system at the seventh, 14 th and 28 th day after intragastric administration respectively. Results: The expressions of b FGF in the lung tissue of the model group enhanced compared with the blank group in the seventh day,14 th day and 28 th day(P〈0.05); the expressions of astragaloside Ⅳ groups and the hormone group weakened obviously compared with the model group over the same period(P〈0.05); the expressions of b FGF in the lung tissue of high and moderate dose groups of astragaloside Ⅳ weakened compared with the hormone group(P〈0.05); there was no difference when low dose group was compared with the hormone group(P〉0.05). Conclusion: Astragaloside Ⅳ could inhibit the expressions of b FGF in the lung tissue of IPF rat model, the intervention effects are positively related to its dose.
