摘要:目的筛选当归、川芎治疗肺纤维化模型大鼠的最佳配比,并观察其对TGF-β1/Smad信号通路的影响。方法 SD大鼠分为8组,即正常对照组,肺纤维化模型组,强的松组,当归组,川芎组,当归、川芎不同配比组,复制肺纤维化大鼠模型并给予相应药物治疗,每天1次,共4周。给药结束后,检测大鼠肺功能,ELISA测定肺泡灌洗液中TNF-α(肿瘤坏死因子-α)、IL-1β(白介素-1β)、IL-6(白介素-6)、NF-κB(核因子-κB)水平。取肺组织测定羟脯氨酸水平,HE、Masson染色后观察其形态。采用免疫组化及RT-PCR技术检测肺组织TGF-β1/Smad信号通路关键分子的表达。结果当归、川芎配伍使用可不同程度改善肺纤维化模型大鼠肺功能,降低炎症因子及羟脯胺酸水平,减轻肺组织病理形态学,以1∶2组作用更明显,并可降低肺纤维化模型大鼠肺组织TGF-β1(转化生长因子-β1)、Smad2、Smad3、α-SMA(α-平滑肌肌动蛋白)的mRNA及蛋白表达。结论当归、川芎1∶2配伍对肺纤维化模型大鼠肺功能有一定改善作用,其机制可能与抑制TGF-β1/Smad信号通路有关。
AIM To screen the best proportion of Angelica sinensis and Ligusticum chuanxiong(Chuanxiong)in treating rat pulmonary fibrosis and to observe their effect in regulating TGF-β1/Smad signaling pathway.METHODS SD rats were randomly divided into 8 groups,and the rat models of pulmonary fibrosis were established.Subsequently,the rats intervened with 4-week corresponding drugs(once daily)were subjected to measurement of lung function,serum oxidative stress indices level and hydroxyproline level in lung tissue;determination of inflammatory factors in alveolar lavage fluid by ELISA;pneumonic assessment of hydroxyproline level;observation of lung morphology by HE and Masson staining;detection of the expression of TGF-β1/Smad signaling pathway key molecules in lung tissues by immunohistochemical method and RT-PCR.RESULTS The combination therapy of Angelica sinensis and Chuanxiong improved the lung function,reduced the levels of inflammatory factors and hydroxyproline acid,and improved the pulmonary morphology of rat models of pulmonary fibrosis.Angelica sinensis and Chuanxiong at ratio of 1∶2 worked best in decreasing the mRNA and protein expression of TGF-β1,Smad2,Smad3 andα-SMA protein in rat models.CONCLUSION Angelica sinensis and Chuanxiong at ratio of 1∶2 can improve the pulmonary function of rats with pulmonary fibrosis,and the therapeutic mechanism may be related to the inhibition of TGF-β1/Smad signaling pathway.
