摘要:目的基于IL-6/STAT3信号通路研究清热排毒胶囊对急性痛风模型大鼠脾脏淋巴细胞增殖的影响。方法将60只大鼠随机分为空白组、模型组、双氯芬酸钠缓释片组,清热排毒胶囊低、中、高组。各组给与相应药物灌胃7天,于第5日灌胃给药1 h后采用MSU溶液踝关节腔注射制备大鼠急性痛风性关节炎模型。各组大鼠于造模后48 h处死并取材。通过酶联免疫吸附法(Elisa)检测各组大鼠踝关节液中IL-6的表达水平;通过CCK8检测脾脏淋巴细胞增值率;通过Western blot检测STAT3及p-STAT3表达水平。结果与模型组比较,清热排毒胶囊各剂量组和戴芬组大鼠踝关节浸出液中IL-6表达水平均显著降低(P<0.05或P<0.01),脾脏淋巴细胞增殖率降低(P<0.05),STAT3、p-STAT3表达水平降低(P<0.05)。结论清热排毒胶囊通过抑制IL-6/STAT3通路激活水平降低大鼠踝关节组织中淋巴细胞增殖来发挥抗炎作用。
Objective To study the effect of Qingre Paidu Capsules on the Splenic Lymphocyte Proliferation in Rats with Acute Gout Based on IL-6/STAT3 Signaling Pathway. Methods Sixty rats were random Ly divided into blank group, model group, diclofenac sodium sustained-release tablet group, and Qingre Paidue Capsule low, middle and high groups. Each group was given intragastric administration of the corresponding drug for 7 days, and on the 5 th day after intragastric administration for 1 hour, an acute gouty arthritis model in rats was prepared by intraarticular injection of MSU solution. The rats in each group were sacrificed 48 hours after the model was established and the materials were taken. The expression of IL-6 in the ankle joint fluid of rats in each group was detected by enzyme-linked immunosorbent assay(Elisa); the spleen lymphocyte proliferation rate was detected by CCK8; and the expression of JAK2 and STAT3 was detected by Western blot. Results Compared with the model group, IL-6 expression in the extracts of the ankle joints of the Qingre Paidiao capsules and the Daifen group was significantly lower(P<0.05 or P<0.01), and the proliferation rate of the spleen lymphocytes was decreased(P<0.05).) STAT3, p-STAT3 expression levels decreased(P<0.05). Conclusion Qingre Paidu Capsule exerts anti-inflammatory effects by inhibiting the activation of IL-6/STAT3 pathway and reducing lymphocyte proliferation in rat ankle joint tissues.
