摘要:目的:比较甘肃产当归不同药用部位对乙酰苯肼所致血虚证干预后的代谢组学差异。方法:将Wistar大鼠按体重随机分为5组:正常对照组、乙酰苯肼模型组、全归给药组、当归身给药组及当归尾给药组(均为10.8 g/kg·d),每组8只,连续给药12 d后腹主动脉取血,用UPLC-Q-TOF/MS检测,并采用代谢组学技术分析血清中内源性代谢产物的变化。结果:与模型组比较,全当归、当归身及当归尾组共有15个内源性代谢物出现回调,全当归与当归身组筛选出7个不同的差异代谢物,全当归和当归尾组筛选出49个差异代谢物。结论:全当归、当归身及当归尾都可能通过嘧啶代谢途径调节代谢通路,全当归与当归尾组可能还调节了饱和脂肪酸代谢、不饱和脂肪酸代谢、糖酵解、胆汁代谢、能量代谢、氨基酸生物合成、谷胱甘肽代谢以及牛磺酸和亚牛磺酸代谢等代谢途径。
Objective: To Compare the metabolomic after the intervention of different medicinal parts of Angelica Sinense on Xuexu Zheng induced by N-acetyl phenyl hydrazine. Methods: Wistar rats were randomly divided into 5 groups: control group,model group,Angelica group,Angelica body and Angelica tail groups. All drugs were administered to rats for consecutive 12 d. The serum sample was detected by UHPLC-QTOF/MS,and the data was analyzed by SIMCA-P14. 0 and Metabo Analyst software,which were adopted by multivariate statistical analysis and univariate statistical analysis. Results: Seven different metabolites were screened between Angelica and Angelica body by SIMCA-P14. 0 and Metabo Analyst software. Metabolites were different between Angelica and Angelica tail,forty nine metabolites were screened. Conclusion: The mechanism of intervention on Xuexu Zheng was similar between Angelica and Angelica body. The metabolomic pathway was regulated by nucleotide metabolism. Compared with Angelica tail group,the mechanism of intervention of Angelica on Xuexu Zheng could be the amino acid metabolism,saturation and unsaturation fatty acid metabolism,galactose metabolism,metabolism of bile,energy metabolism,glutathione metabolism,taurine and sulfonic acid metabolism.
