摘要:目的比较性状鉴别与灰色关联法对不同规格大黄饮片质量评价的差异性。方法收集不同产地10种不同规格大黄饮片,基于“辨状论质”性状鉴别法和“德尔菲法”建立性状鉴别特征质量评价表,参照《中华人民共和国药典(一部)》2020年版分别测定样品的水分、总灰分、浸出物、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚等8个指标的含量。结果性状鉴别特征质量评分和灰色关联法的质量排序比较,甘南(头黄)和甘南(头片)均排名靠前,断面颜色较深,有较明显的槟榔茬和星点等特征,质量最优,与饮片市场中的商品等级划分一致;甘南(饮片,直径>2.5 cm)和青海湟中县(大黄片)均排名靠后,表面特征无锦纹,且颜色较浅,品质较劣。2种评价方法的排序结果除R3、R5、R10样品的排序有较小差异外,其他排名相差不大。结论新构建的大黄性状鉴别特征量化质量评价表具有各评价指标主次分明、使用简单、易于推广等特点,与灰色关联法评价样品实际质量的契合度较高。
Objective To compare the differences between macroscopic identification and grey relational analysis(GRA)in quality assessment of Rhei radix et rhizoma decoction pieces with different specifications.Methods Ten different specifications of Rhei radix et rhizoma decoction pieces from different producing areas were collected,the quality assessment scale of macroscopic identification was established based on the method of“quality assessment based on feature identification”and“Delphi method”.The content of 8 indices,namely water,total ash,extract,aloe emodin,rhein,emodin,chrysophanol and emodin methyl ether etc.was determined by reference to Pharmacopoeia of the People’s Republic of China(2020 Edition).Results Comparing the quality score of macroscopic identification with the quality ranking of GRA,Gannan(touhuang)and Gannan(toupian)rank high,the section color is darker,and there are obvious features such as areca stubble and star points.The quality is the best,which is consistent with the commodity grade classification in the decoction piece market;Gannan(decoction pieces,diameter>2.5 cm)and Huangzhong County of Qinghai Province(Dahuang pian)rank lower,with no brocade pattern on the surface,light color and poor quality.The ranking results of the 2 assessment methods have little differences except for R3,R5 and R10 samples.Conclusion The newly constructed quantitative quality assessment scale of macroscopic identification of Rhei radix et rhizoma has the characteristics of distinct primary and secondary assessment indexes,simple use and easy promotion,which is highly consistent with the actual quality assessment of samples based on the GRA.
