摘要:目的研究逍遥散及其配伍组方对氢溴酸东莨菪碱所致阿尔茨海默病小鼠学习记忆、抗氧化及中枢神经递质活性的影响,探讨逍遥散防治阿尔茨海默病的作用及其机制,阐释疏肝、养血、健脾相结合的配伍机制。方法 60只SPF级KM小鼠随机分为空白组、模型组、逍遥散全方组、疏肝养血组、疏肝健脾组、健脾养血组等6组;采用腹腔注射氢溴酸东莨菪碱复制阿尔茨海默病小鼠;测定小鼠学习记忆能力及脑组织匀浆SOD、MDA、AchE、ChAT及MAO活性。结果逍遥散全方组、疏肝养血组、疏肝健脾组、健脾养血组均可提高小鼠学习记忆能力(P<0.05或P<0.01),降低潜伏时间(P<0.05或P<0.01);可提高脑组织匀浆SOD、AchE及MAO活性(P<0.05或P<0.01),降低MDA、ChAT活性(P<0.05或P<0.01)。但学习记忆、抗氧化及神经递质活性的改善皆以逍遥散全方组最为明显(P<0.05)。结论逍遥散能较好改善阿尔茨海默病小鼠学习记忆、抗氧化及中枢神经递质活性,对阿尔茨海默病具有较好的防治作用,提示疏肝、养血、健脾相结合的配伍思路用于阿尔茨海默病的防治是科学合理的。
Objective To observe the effect of Xiaoyao powder and it's splited prescription on the ability of learning and memory of Alzheimer's disease(AD) model mice made by scopolamine hydrobromide and explain mechanism of incorporation of Shugan, Yangxue and Jianpi. Methods 60 SPF KM mice were randomly divided into blank control, model and all party, Shugan Jianpi, Shugan Yangxue and Jianpi Yangxue groups of Xiaoyao powder. AD mice model was induced with scopolamine hydrobromide alkali intraperitoneal injection. The abilities of learning and memory and the levels of SOD, MDA, AchE, ChAT and MAO in brain homogenate were measured. Results In all groups, Shugan Jianpi, Shugan Yangxue, Jianpi Yangxue groups of Xiaoyao powder, the abilities of learning and memory were improved and latency were decreased and the levels of SOD, AchE, MAO, were increased, the levels of MDA, ChAT were decreased, and all groups of Xiaoyao powder were most obviously. Conclusions Xiaoyao powder could better improve the abilities of learning and memory and antioxidant and the activity of central neurotransmitter of AD model mice, show that compatibility ideas of incorporation of Shugan, Yangxue and Jianpi are scientific and reasonable idea on preventing from AD.
