摘要:目的通过检测甘南地区部分体检者血清中TTV,分析TTV在该地区的流行情况及特征。方法随机收集甘南州人民医院藏族体检者血清362份,巢式聚合酶链式反应检测血清中TTV-DNA;抽取8份阳性血清扩增产物测序并进行同源性比较和系统进化分析。结果 362份血清中,阳性血清128份(男性81份、女性47份),阳性率为35.4%(128/362),男性与女性之间TTV感染差异具有统计学意义(P<0.05)。序列同源性分析表明,8份阳性血清序列与日本参考株、北京株和深圳株对应核苷酸序列的同源性在88.7%-100%;分子进化树分析显示,8个序列分布在2个不同的分支中,7个序列与北京株、日本参考株TA278同属1个分支,1个序列与深圳株组成另1个分支,且其同源性为99.9%,提示其为同一型或亚型。结论这一结果表明在甘南藏族人群中存在TTV感染,将为预防甘南地区TTV流行及其与其他疾病的相关性研究奠定基础。
Objective To detect existing TTV in the serum from physical examination of Tibetan people in Gannan regions,and to analyze the prevalence and characteristics of TTV in the region. Methods 362 serum were randomly collected from physical examination of some tibetan people in Gannan regions. TTV-DNA extracted from the serum was examined by the nested polymerase chain reaction. 8 amplicons from positive serum were sequenced,compared,and analyzed by Blast,Clustal X and MEGA softwares. Results Of the 362 serum samples,128( 81 males and 47 females) were tested positive for TTV at a rate of 35. 4%( 182 /362). The significant difference of TTV infection was found between males and females.Sequence analysis showed that the sequence homologies among the TTV isolates from 8 positive serum in Japan,Beijing and Shenzhen ranged from 88.. 7% to 100%. 8 TTV sequences were divided into two branches by phylogenetic analysis. 7 sequences belonged to one branch with Beijing strain and Japanese reference strain TA278,and one sequence,whose homology was 99. 9% with Shenzhen strain,belonged to another branch,suggesting that the isolates belonged to the same type or subtype. Conclusion TTV infection widely exists in Tibetan people. It will lay the foundation for the prevention of the TTV epidemic and future research on its correlation with other diseases in Gannan area.
