摘要:目的观察红芪多糖(HPS)对糖尿病周围神经病变(ob/ob)小鼠坐骨神经中丝裂原活化蛋白激酶(MAPK)信号通路的影响。方法将50只ob/ob小鼠按体重随机分为模型组、低、中、高剂量实验组和对照组,每组10只;另取10只C57BL/6小鼠为正常组。正常组和模型组均给予5 mL·kg^(-1)·d^(-1)蒸馏水;对照组按30 mg·kg^(-1)·d^(-1)的剂量给予0.04 mg·mL^(-1)α-硫辛酸;低、中、高剂量实验组分别按50,100和200 mg·kg^(-1)·d^(-1)的剂量给予20 mg·mL^(-1)HPS。6组小鼠每天灌胃给药1次,连续给药8周。用Power Lab生理检测仪检测小鼠的运动神经传导速度(MNCV),用蛋白质印迹法测定坐骨神经组织中磷酸化p38 MAPK(p-p38MAPK)和白细胞介素-6(IL-6)蛋白的表达水平。结果治疗8周后,低、高剂量实验组和对照组、模型组、正常组的MNCV分别为(40.35±2.12),(43.74±3.21),(42.70±3.36),(40.29±1.79)和(46.95±2.36)m·s^(-1),p-p38MAPK/p38MAPK蛋白相对表达量分别为1.03±0.02,0.77±0.05,0.86±0.06,1.20±0.03和0.63±0.04,IL-6蛋白相对表达量分别为0.77±0.03,0.67±0.01,0.72±0.01,0.87±0.02和0.56±0.03。模型组的上述指标与正常组和高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论HPS可通过下调p-p38MAPK/p38MAPK及IL-6的表达,从而改善对糖尿病周围神经病变的炎症反应。
Objective To observe the effect of Hedysarum polybotrys polysaccharides(HPS)on the signaling pathway of mitogen-activating protease(MAPK)in the sciatic verve of diabetic peripheral neuropathy(ob/ob)mice.Methods Fifty ob/ob mice were randomly divided into model group,experimental-L,-M,-H groups and control group according to body weight,with 10 mice in each group.Another 10 C57 BL/6 mice were selected as normal group.Normal group and model group were given 5 mL·kg^(-1)·d^(-1)distilled water.The control group was given 30 mg·kg^(-1)·d^(-1)alpha-lipoic at the concentration of 0.04 mg·mL^(-1).The experimental-L,-M,-H groups were given 50,100 and 200 mg·kg^(-1)·d^(-1)HPS at a concentration of 20 mg·mL^(-1),respectively.Six groups were given intragastric administration once a day for 8 weeks.The motor nerve conduction velocity(MNCV)of mice was measured by Power Lab physiological analyzer.The expression levels of phosphorylated p38 mitogen-activating protease(p-p38 MAPK)and interleukin-6(IL-6)in sciatic nerve were determined by Western blot.Results After 8 weeks of treatment,the MNCV of experimental-L,-H groups and control group,model group and normal group were(40.35±2.12),(43.74±3.21),(42.70±3.36),(40.29±1.79)and(46.95±2.36)m·s-1,the relative expression levels p-p38 MAPK/p38 MAPK proteins were1.03±0.02,0.77±0.05,0.86±0.06,1.20±0.03 and 0.63±0.04,the relative expression levels of IL-6 protein were 0.77±0.03,0.67±0.01,0.72±0.01,0.87±0.02 and 0.56±0.03,respectively.Compared with the normal group and the experimental-H group,the above indexes in the model group had statistical significance(all P<0.05).Conclusion HPS can improve the inflammatory response to diabetic peripheral neuropathy by down-regulating the expression of p-p38 MAPK/p38 MAPK and IL-6.
