摘要:基于HPLC法与正交试验优化苦参中苦参碱的最佳提取工艺。采用正交试验选取氨水用量A(倍)、氯仿用量B(倍)、提取时间C(h)3个因素,并基于优化的色谱条件进行苦参碱含量检测。结果显示,正交试验苦参碱提取工艺的最佳组合为A_(2)B_(1)C_(1),即40倍氯仿量、2倍氨水量、回流提取3次,每次提取1 h。说明该试验所得的苦参碱提取工艺简单可行,为苦参碱的基础应用提供一定参考。
The optimal extraction process of matrine in Sophora flavescens was optimized based on HPLC method and orthogonal experiment.Orthogonal experiment was used to select three factors:ammonia dosage,chloroform dosage,and extraction time,and the matrine content was detected based on optimized chromatographic conditions.The results showed that the best combination of matrine extraction process in orthogonal test was A_(2)B_(1)C_(1),which was 40 times the amount of chloroform,2 times the amount of ammonia,and refluxed for 3 times,each extraction for 1 hour.It showed that the extraction process of matrine obtained in this experiment was simple and feasible,and provided a certain reference for the basic application of matrine.
