摘要:目的通过系统研究将超滤技术应用于红芪提取液的纯化工艺。方法用水提取红芪、离心分离得滤液。以总黄酮、总多糖、正丁醇浸出物为判据,以膜分子截留量、工作压力、药液温度、浓缩比为参数的均匀设计来优化超滤工艺参数。结果优化所得红芪提取液超滤工艺为用分子截留量10万的膜,在压强0.07 MPa,药液温度25℃,浓缩程度1∶18的条件下超滤,超滤膜表面先用5 g/L的tween-20处理5 min。结论优化所得超滤工艺与未超滤相比有较高的红芪总黄酮和总多糖得率。
AIM To apply the ultrafiltration to the purification process of the extract of Hedysarum polybotrys.METHODS Hedysarum polybotrys was extracted by drinking water,and then centrifuged to get the extract.Total flavones,total polysaccharides,and n-butanol extract were adopted as the markers,molecular weight in cake,pressure,suspension temperature,and concentration ratio as parameters for even design.Extraction parameters of extract was evaluated and optimized.RESULTS The experiment showed that the optimized ultrafiltration process was: the ultrafiltration pressure(0.07 MPa),the extraction temperature(25 ℃) and the concentration ratio(1 ∶ 18),using ultrafiltration membrane of MWCO(ten thousand molecular weight),pretreated with tween-20(5 g/L) for 5 min.CONCLUSION The optimized ultrafiltration process can obtain the higher yield of total flavones and total polysaccharides as compared with the yield of non-ultrafiltration of Hedysarum polybotrys.
