摘要:目的采用正交试验法优选黄芪水提液提取的最佳条件,利用Box-Behnken响应面法优化黄芪及药物配比研究。方法水浴回流法提取黄芪,以干膏率为评价指标,在料液比、提取时间、提取次数3个单因素的基础上进行正交试验优选黄芪水提液的最佳提取条件。该实验以黄芪为原料,在单因素的基础上,以黄芪浓缩液/药物混合液、辅料1加入量、辅料2加入量、柠檬酸加入量为考察因素,优化黄芪及药物最佳配比。结果黄芪水提液最佳提取条件为料液比1∶6,提取时间1 h,提取次数3次,黄芪的提取率为44.75%。黄芪及药物配比为以50 g药物混合液为基准,黄芪浓缩液/药物混合液为29%,辅料1加入量0.15 g,辅料2加入量20 g,柠檬酸加入量0.2 g,最终感官评分达92.5分,与预测值相近,说明模型拟合良好,该优化工艺准确可行。结论黄芪最佳提取工艺及其药物配比优化结果重现性好,工艺简便易行,可以为黄芪系列产品的开发奠定基础,有一定的开发应用前景。
Objective To use the orthogonal test method,the best conditions for extracting the extraction extraction extraction of Huangqi(Astragali Radix),using the Box-Behnken response surface method to optimize the study of Huangqi(Astragali Radix)and drug ratio.Methods The water bath retraction method extracts Huangqi(Astragali Radix),and the orthogonal test is preferably the optimal extraction conditions for the orthogonal test by three single factors based on the liquid ratio,extraction time,and the number of extracted times.Similarly,in this experiment,Huangqi(Astragali Radix)was used as raw material,on the basis of single factor,Huangqi(Astragali Radix)concentrate/drug mixture,excipient 1,excipient 2 and citric acid were used as factors to optimize the ratio of Huangqi(Astragali Radix)and drug by Box-Behnken response surface methodology.Results The optimal extraction conditions of Huangqi(Astragali Radix)were as follows:solid-liquid ratio 1∶6,extraction time 1 h,extraction times 3 times,the extraction rate of Huangqi(Astragali Radix)was 44.75%.The ratio of Huangqi(Astragali Radix)to medicine was based on 50 g drug mixture,29%Huangqi(Astragali Radix)concentrate/medicine mixture,0.15 g excipient 1,20 g excipient 2 and 0.2 g citric acid,and the final sensory score was 92.5 points,which was close to the predicted value,indicating that the model fit well and the optimization process was accurate and feasible.Conclusion The optimal extraction process of Huangqi(Astragali Radix)and its drug ratio optimization results were reproducible and simple,which could lay a foundation for the development of Huangqi(Astragali Radix)series products and had a certain prospect of development and application.
