摘要:目的利用生物信息学方法分析烧伤创面相关基因及生肌玉红膏治疗烧伤创面的作用靶点。方法从基因表达数据库(GEO)中下载烧伤相关数据集,将样本分为4组:正常组和烧伤后0~3、4~7、>7 d组。筛选烧伤后创面组织差异表达基因(DEGs)并对其进行基因本体(GO)及通路富集分析。构建蛋白互作(PPI)网络并筛选关键基因。检索生肌玉红膏各单味药(当归、白芷、紫草、血竭和甘草)作用靶点。结果3个烧伤后不同时间组与正常组比较,DEGs主要与中性粒细胞活化、炎症反应及免疫反应等有关,且富集在细胞因子-细胞因子受体、肿瘤坏死因子(TNF)等通路。蛋白互作分析获取白细胞介素-6(IL-6)、趋化因子配体8(CXCL8)和基质金属蛋白酶9(MMP9)等11个关键基因。生肌玉红膏各单味药作用靶点中包含有IL-6、MMP9等烧伤创面关键基因。结论IL-6、CXCL8和MMP9等基因在烧伤创面损伤中发挥重要作用,生肌玉红膏可能通过影响这些关键基因对烧伤创面发挥治疗作用。
Objective To analyze the related genes of burn wound by bioinformatics and screen the target of Shengji Yuhong ointment in treating burn wounds.Methods Burn related dataset was downloaded from Gene expression database(GEO)and divided into four groups:normal group,0-3 d after burn group,4-7 d group,and>7 d group.Differentially expressed genes(DEGs)were screened and analyzed for gene ontology(GO)and pathway enrichment.Protein protein interaction(PPI)network was constructed and key genes were screened.To search the target of Shengji yuhong ointment.Results Comparison between three different time burn groups and normal group,DEGs were mainly related to neutrophil activation,inflammatory response and immune response,and enriched in cytokines receptor,tumor necrosis factor(TNF)and other pathways.Eleven key genes such as interleukin-6(IL-6),Chemokine ligand 8(CXCL8)and matrix metalloproteinase9(MMP9)were obtained by protein interaction analysis.The key genes of burn wound such as IL-6 and MMP9 were included in the target of Shengji Yuhong ointment.Conclusion CXCL8,MMP9,IL-6 and other genes play an important role in burn wound injury.Shengji Yuhong ointment may play a therapeutic role in burn wound by affecting some key genes.
