摘要:目的:研究不同剂量葛根素对去卵巢大鼠学习记忆能力和海马结构的影响。方法:SD雌性大鼠,切除双侧卵巢后,连续腹腔注射葛根素(120,60,30mg/kg)30d后,用Morris水迷宫检测大鼠的学习记忆能力;通过电镜观测与学习记忆相关脑区海马CA1的突触超微结构变化。结果:葛根素高剂量(120mg/kg)和中剂量(60mg/kg)组大鼠逃避潜伏期随着训练天数的增加明显缩短(P<0.01,P<0.05),倍美力片(120mg/kg)、葛根素高剂量(120mg/kg)和中剂量(60mg/kg)组大鼠跨越平台次数明显增加(P<0.05),且在平台象限停留时间均明显延长(P<0.05);模型组大鼠海马CA1区神经细胞核明显固缩,染色质浓缩成团,分布于核膜边缘,核仁不清楚,核膜增厚,部分断裂,核内可见空腔,胞浆内大量脂褐素堆积,而葛根素高剂量(120mg/kg)组则细胞核形态较规则,染色质分布均匀,核仁明显,核膜光滑;葛根素中剂量(60mg/kg)组细胞核轻度浓缩,染色质分布较均匀,核内偶见空腔,胞浆内少量脂褐素堆积;但葛根素低剂量(30mg/kg)组与模型组相似,细胞核形态相对较规则。结论:葛根素对去卵巢大鼠学习记忆能力减退和海马CA1结构异变具有一定的保护作用。
Objective:To study the effects of Puerarin on learning and memory ability and hippocampal structure in ovariectomized rats.Methods:The female rats were randomly assigned into six groups:model group,premarin group,Puerarin groups(120,60,30mg/kg),and control group,continuous treatment 30d,The rats' learning and memory ability was tested in Morris maze,and the hippocampal CA1 synaptic ultrastructure was observed by electron microscope.Results:The rats' escape latencies was shortened with increasing training days in high-dose(120mg/kg) and mid-dose(60mg/kg) Puerarin(P0.01,P0.05),the times across the platform was increased in high-dose(120mg/kg) and mid-dose(60mg/kg) Puerarin groups(P0.05),and the quadrant dwell time in the platform were prolonged also(P0.05);The model group rats' nerve nuclei were destroied,the nucleolus was not clear,the cytoplasm was abnormal in area CA1 of the hippocampus,and the rats nuclear shape is regular,and nuclear membrane is smooth in high dose(120mg/kg) puerarin;the rats nuclei morphology and cytoplasmic lipofuscin is improved obviously in mid-dose(60mg/kg) puerarin group,but the low-doses(30mg/kg) group is worse.Conclusion:The learning and memory deficits or hippocampal CA1 structure destroyed have been protected effectively by Puerarins in ovariectomized rats.
