摘要:目的探讨芒见赛保总萜对H_(22)肝癌细胞移植瘤小鼠Th1/Th2漂移及Bax/Bcl-2表达的影响。方法昆明种小鼠皮下接种H_(22)肝癌腹水建立荷瘤动物模型,观测小鼠的一般状况、体质量变化、抑瘤率及脏器指数,ELISA法检测血清及脾脏组织中Th1(IL-2、IFN-γ)及Th2(IL-4、IL-10)型细胞因子水平,免疫组化法检测肿瘤组织中Bax和Bcl-2蛋白表达水平,HE染色观察肿瘤组织及肝脏的病理变化。结果 100~400 mg/kg芒见赛保总萜可明显改善H_(22)肝癌移植瘤小鼠一般状况,抑制肿瘤生长,升高血清及脾脏IL-2水平,降低IFN-γ、IL-4、IL-10、IFN-γ/IL-4比值,改善Th1/Th2漂移状态,增强肿瘤组织中Bax表达,并抑制Bcl-2表达从而升高Bax/Bcl-2比值,缓解肿瘤及肝脏组织的病理学变化。结论芒见赛保总萜可明显抑制小鼠H_(22)肝癌的生长,改善小鼠血清及脾脏Th1/Th2漂移,提高肿瘤组织Bax/Bcl-2比值可能是其抗肿瘤的作用机制之一。
This study was performed to investigate the influence of total terpenes extract of Cremanthodium humile Maxim (TTC) on Th1/Th2 and Bax/Bcl-2 balance in H22 tumor-bearing mice. The mouse models of H22 liver cancer were established by inoculation of H22 ascites. And then the general condition and the body weight change of mice were observed, the tumor inhibition rate and organ index of each group were calculated. Enzyme-linked immunosorbent assay (ELISA) was used to measure the levels of Thl (IL-2, IFN-γ) and Th2 (IL-4, IL-10) in serum and spleen of mice; immunohistochemical method was used to detect the expression of Bax and Bcl-2 proteins in tumor tissu staining. Data showed that 100-400 es. The pathological changes of tumor and liver tissue were observed by HE mg/kg TTC could significantly improve the general condition and inhibite tumor growth in H22 tumor-bearing mice. The level of IL-2 in serum and spleen were increased, but the levels of IFN-γ, IL-4, IL-10, IFN-γ/IL-4 in serum and spleen were decreased in TTC treated mice, while Th1/ Th2 drift state was inhibited by Immunohistochemical down-regulate up-regulate staining showed that TTC expression expression of Bax, of while Bcl-2 TTC. could and HE stainingshowed that TTC could improve the pathological changes of tumor and liver tissues. In conclusion, TTC can inhibit tumor growth, and the mechanism may relate to inhibiting the Thl/Th2 drift state, and increasing the ratio of Bax/Bcl-2 in tumor tissue.
