摘要:目的 探索玻璃化超低温过程中不同预处理和PVS2处理时间两个关键环节对留兰香再生植株遗传稳定性的影响,为植物种质资源超低温保存提供理论依据。方法 使用改良CTAB法提取留兰香超低温再生植株DNA,用16条引物分别对不同预处理和不同PVS2处理后的35株留兰香超低温再生植株DNA进行ISSR-PCR扩增检测。结果 不同浓度蔗糖和预处理时间,共扩增出3465条带,均未检测到变异条带;不同PVS2处理时间,共扩增出条带1776条,也未出现变异条带。形态学指标测定结果表明,超低温保存再生植株的株高、茎粗、主茎叶片数、节间数、叶片色及叶片数均未发生变异。结论 一定范围内高浓度蔗糖预处理和不同PVS2处理时间不会引起超低温保存再生植株产生变异,留兰香的超低温再生植株具有遗传稳定性,玻璃化超低温保存可作为一种留兰香优良种质资源保存的方法。
Objective To explore the effects of different pretreatments and PVS2 treatment time on the genetic stability of the regenerated plants of Mentha spicata L.,and to provide theoretical basis for the cryopreservation of plant germplasm resources.Methods Total genomic DNA was extracted from the regenerated plants of the Mentha spicata L.by the CTAB method.ISSR-PCR amplification assay was performed on 96 plants of cryopreservation regenerated plants originated from different pretreatments and PVS2 treatments with 16 primers.Results A total of 3465 scored bands were generated from different concentrations of sucrose and pretreatment times,and no polymorphic bands were detected;1776 bands were obtained from regenerated plants of different PVS2 treatment times,and no polymorphic bands appeared either.The results of morphological indexes showed that there was no variation in plant height,stem thickness,number of main stem leaves,number of internodes,leaf color and number of leaves of the regenerated plants preserved at cryopreservation.Conclusion The high concentration of sucrose pretreatment and different PVS2 treatment time within a certain range did not cause mutations in the cryopreservation preserved regenerated plants,and the cryopreservation regenerated plants of Mentha spicata L.had genetic stability,and vitrification cryopreservation preservation could be used as a method to preserve excellent germplasm resources of Mentha spicata L.
