摘要:目的研究当归挥发油对自发性高血压大鼠(SHR)肝脏组织形态学的影响及肝脏脂质代谢相关基因肿瘤坏死因子α诱导8样分子2(Tnfaip8l2)、血清胎球蛋白A(Ahsg)表达的影响,探讨当归挥发油对SHR脂代谢的影响及其机制。方法将24只8周龄雄性SHR随机分为模型组、当归挥发油组和阿托伐他汀钙组,并用同周龄的Wistar大鼠作为对照组。每周监测各组大鼠血压。分别灌胃4 w后,取大鼠肝脏组织,HE染色观察肝脏组织形态学的变化,免疫印迹法检测Tnfaip8l2、Ahsg蛋白水平的表达。qRT-PCR检测Tnfaip8l2、Ahsg mRNA的表达。结果 HE染色显示,与模型组比较,当归挥发油组病变有所减轻,阿托伐他汀钙组未见明显异常。与模型组比较,Tnfaip8l2、Ahsg蛋白在各用药组表达上调。结论当归挥发油对肝脏组织有保护作用,并且能降低SHR的血压。当归挥发油对SHR血脂有调节作用。当归挥发油可能通过上调Tnfaip8l2、Ahsg基因的表达水平、抑制动脉粥样硬化而降压,并且影响脂代谢。
Objective To study the effect of Chinese angelica naphtha on liver lipid metabolism related Tnfaip8 l2, Ahsg gene expression in spontaneously hypertensive rats(SHRs). Methods 24 male 8-week-old SHRs were randomly divided into model group, angelica naphtha group and atorvastatin calcium group. Wistar rats, as old as SHR were selected into normal control group. Blood pressure was monitored weekly. After 4 weeks, the liver tissues of rats were acquired to observe the change of liver tissue morphology by HE staining. Western blot was used to detect Tnfaip8 l2,Ahsg protein expression. qRT-PCR was used to detect Tnfaip8 l2,Ahsg mRNA expression.Results HE staining showed that compared with model group, lesions of liver were decreased in angelica naphtha group and there were no obvious abnormal lesions in atorvastatin calcium group. Western blot showed that compared with model group, Tnfaip8 l2,Ahsg protein expressions in each treatment group were up-regulated. Conclusions Angelica volatile oil reduces SHR′s blood pressure. Tnfaip8 l2 and Ahsg are up-regulated and differentiated in each drug group. Angelica volatile oil has regulatory effect on SHR fat metabolism, which may increase the mRNA level of Tnfaip8 l2 and Ahsg gene, inhibit atherosclerosis, lower the blood pressure of and affect lipid metabolism.
