摘要:骨质疏松症(osteoporosis,OP)是中老年人群最常见的骨骼疾病,随着我国老龄化社会的到来,OP已严重影响国人生活质量,现阶段对OP的治疗已经有了相对系统的方案,但抗骨质疏松是一个长期的过程,药物使用周期长、费用高、不良反应较多等一系列问题都为我国的医疗带来了巨大的压力,所以继续研究OP发病机理,寻求更有优势的抗骨质疏松药物显得尤为重要。现研究发现肿瘤坏死因子α(tumor necrosis factorα,TNF-α)在OP进程中起关键作用,TNF-α介导的骨组织细胞直接或间接发生坏死是导致骨量减少的一个关键因素,TNF家族作为免疫因子对抵抗致病因素对机体的损害起了巨大作用,但是机体内过多的TNF-α则会诱导某些疾病的发生,如OP,它参与多种骨组织细胞死亡方式。本文综述了TNF-α通过程序性坏死、铁死亡、细胞凋亡、自噬等程序性死亡方式导致OP的相关通路,为OP的治疗提供理论依据。
Osteoporosis(OP)is the most common bone disease in the elderly population,and with the aging of population,OP seriously reduces the quality of life of Chinese people.At present stage there is a relatively systematic program for the treatment of OP,but anti-osteoporosis is a long-term process,and there are some difficulties such as long usage cycle of drug,high cost and significant side effects which bring tremendous pressure to healthcare in our country.It is particularly important to continue to study the pathogenesis of OP and seek more advantageous anti-osteoporotic drugs.Current studies found that tumor necrosis factorα(TNF-α)played a key role in the process of OP,and direct or indirect necrosis of osteocytes mediated by TNF-α was a key factor leading to bone loss.TNF family as an immune factor has a great effect on the resistance to the pathogenic factors that damage to the body.However,excessive TNF-α in the body will induce the occurrence of certain diseases like OP.TNF-α involves in various modes of cell death in bone.In this article,the related pathways of OP caused by TNF-α through programmed necrosis,ferroptosis,apoptosis,autophagy and other programmed death modes were reviewed,so as to provide a basis for the treatment of OP.
