摘要:目的:对甘肃、陕西、宁夏、四川及青海5个主产地秦艽的遗传多样性进行分析。方法:用随机扩增多态性DNA标记(RAPD)技术对11个秦艽种群90个个体基因组DNA的遗传多样性进行检测。结果:RAPD 8条引物检测到65条扩增条带,其中有59条是多态的,多态位点百分比达到97.78%。通过聚类分析可将秦艽的11个种群聚为两类,四川和陕西两省4个种群聚为一类;甘肃、宁夏和青海省7个种群聚为一类。结论:甘肃省环县种群、子午岭种群和华亭种群应作为秦艽的重要种群加以优先重点保护。
Objective:The study analyzed the genetic diversity of G.macrophylla Pall.in its native growth places in Gansu,Shanxi,Ningxia,Sichuan and Qinghai provinces.Methods: Detect the genetic diversity of the DNA of 90 G.macrophylla Palls from 11 populations through RAPD technique.Results: 65 expansion bands with 59 being polymorphism were detected through 8 primers.Polymorphism rate is 97.78%.The 11 populations were classified into 2 groups through classification analyses,with 4 populations from Sichuan and Shanxi provinces into one group,and 7 populations from Ganshu,Ningxia and Qinghai provinces into another group.Conclusion: The populations in Huan,Ziwuling,and Huating Counties of Gansu Province should be protected with priority as the important populations.
