摘要:目的:观察"疏肝调神"针法对创伤后应激障碍(PTSD)模型大鼠海马CA1、CA3区异常神经编码时空模式的影响。方法:50只SD大鼠随机分为空白组、模型组、抓取组、西药组和针刺组,每组10只。除空白组外,其余组以复合应激法造模同时对应组分别以盐酸帕罗西汀、"疏肝调神"针法干预和抓取固定治疗。在体多通道记录海马CA1、CA3区神经元集群电活动,分析放电频率和波形幅值,留取图谱。结果:与空白组相比,模型组和抓取组CA1、CA3区放电频率缩短,集中分布频带降低,呈阵发性;放电波稀疏、凌乱,波形变窄,波形幅值减小(P<0.01)。与抓取组相比,西药组和针刺组CA1、CA3区放电频率延长,集中分布频带升高,呈断续性;放电波规则、整齐,波形幅值增大(P<0.01);两组CA1区波形增宽,但CA3区仅针刺组增宽。与西药组相比,针刺组CA3区波形幅值较宽,平均波形幅值较高(P<0.05)。结论:复合应激法可使大鼠海马CA1、CA3区神经编码时空模式异常改变,相关治疗均可显著调节部分异常改变,但"疏肝调神"针法整体效应更明显,这可能是针刺干预PTSD的重要中枢机制。
Objective: To investigate the effect of 'dispersing liver and regulating spirit' acupuncture therapy on spatial and temporal patterns of neural coding in hippocampal CA1 and CA3 regions of rats with PTSD. Methods: Fifty SD rats were divided randomly into 5 groups: control group, model group, grasped group, acupuncture group and paroxetine hydrochloride group(10 rats in each group). Except for control group, all the other rats were stimulated by the composite stress method to duplicate the PTSD model, meanwhile the paroxetine hydrochloride group was intervened by the gastric perfusion of paroxetine hydrochloride, the acupuncture group was intervened by 'dispersing liver and regulating spirit' acupuncture treatment, the grasped group was grasped by the same way as the acupuncture group and the paroxetine hydrochloride group were fastened. After the intervention, neuron cluster electrical activity of hippocampal CA1 and CA3 regions was recorded by in vivo multi-channel recording methods, the discharge frequency and the wave amplitude were analyzed, and the relative spectrums were drawn. Results: Compared to the control group, the model group and the grasped group showed the shortening of discharge frequency in CA1 and CA3 regions, and the concentrated band was reduced and paroxysmal, the discharge wave was sparse and messy, the waveform was narrowing and its amplitude was reducing, which all showed significant difference(P〈0.01). Compared to the grasp group, the paroxetine hydrochloride group and the acupuncture group showed increase in pike quantity of action potential, and the prolonged discharge frequency, and the concentrated band was reduced and paroxysmal, the discharge wave was orderly and regulated, the waveform amplitude was amplified, which all showed significant difference(P〈0.01). The waveform of both group showed broadening in CA1 region but only the acupuncture group showed the similar change in CA3 region. Compared to the paroxetine hydrochloride group, the acupuncture group showed significantly broadening in the waveform CA3 region and higher average amplitude of waveform(P〈0.05). Conclusion: The stimulation from the composite stress method could cause the abnormality of the spatiotemporal pattern of hippocampal CA1, CA3 regions, and related treatment could significantly adjust some abnormal changes. However, compared to the other treatments, 'dispersing liver and regulating spirit' acupuncture method has a more significant therapeutic effect, which is important central mechanism of treating PTSD by acupuncture therapy.
