摘要:BACKGROUND: This work aimed to further explore the medication rules of Chinese medicine in treating insomnia. METHODS: A total of 104 prescriptions were statistically analyzed. The association rule algorithm was applied to mine the core drug combinations, network pharmacology was applied to analyze the core drug combinations’ mechanism of action, and molecular docking was applied for validation.RESULTS: The most frequently used drugs in prescriptions for treating insomnia are Semen Ziziphi Spinosae, Radix Glycyrrhizae, Ginseng, and Indian Bread With Pine. The majority of the drugs utilized were those that supplementing drug, followed by drugs that heat-clearing, mind-caming, and drugs that exterior-releasing. These drugs have properties of warmth and coldness and flavors of sweetness, spiciness, and bitterness. They affect the meridians of the liver, lungs, spleen, kidneys, heart, and stomach. The association rules were applied to analyze the core drug combinations, which included Radix Astragali–Ginseng, Rhizoma Chuanxiong–Radix Glycyrrhizae, Seman Platycladi–Semen Ziziphi Spinosae, Pericarpium Citri Reticulatae–Radix Glycyrrhizae, Radix Polygalae–Semen Ziziphi Spinosae, and Radix Astragali–Semen Ziziphi Spinosae. Network pharmacology was used to analyze the drug pair of Radix Polygalae–Semen Ziziphi Spinosae in the treatment of insomnia through the cAMP signaling pathway, which involved multiple components and targets. Molecular docking revealed that the drug pair components showed excellent binding to targets.CONCLUSION: Chinese medicine effectively treats insomnia by utilizing a combination of medications that target multi-ingredients, multi-targets, and multi-signaling pathways. ? 2024, The Authors.
