摘要:目的:探讨不同负荷运动对大鼠心肌线粒体电子传递链功能的影响。方法:将50只雄性Wistar大鼠随机分为5组(n=10):安静对照组(C)、低负荷运动组(LE)、中等负荷运动组(ME)、高负荷运动组(HE)和极高负荷运动组(SHE)。各运动组按不同强度运动训练方案进行8周递增负荷跑台运动,每周运动6d,1次/d。运动训练结束后,取大鼠心脏样本,用差速离心法提取线粒体并测定线粒体电子传递链复合体酶——NADH-Q还原酶、琥珀酸-Q还原酶、细胞色素还原酶和细胞色素氧化酶的活性。结果:与C组相比,ME和SHE组的NADH-Q还原酶活性均显著升高(P<0.01),HE组显著下降(P<0.05);ME组的琥珀酸-Q还原酶活性显著升高(P<0.05),HE组显著下降(P<0.01);ME和SHE组的细胞色素还原酶活性均显著升高(P<0.01);ME、HE和SHE组的细胞色素氧化酶活性均显著升高(P<0.05,P<0.01)。结论:不同负荷的运动对大鼠心肌线粒体电子传递链复合体酶活性均有影响,在提高心肌线粒体电子传递链功能方面,中等负荷运动效果最佳,其次为极高负荷运动。
Objective:It is to investigate the effects of training with different load on the electron transport chain in mitochondrial of myocardial in rats.Methods:Fifty male Wistar rats were randomly divided into 5 groups(n=10):control(C),low load exercise(LE),medium load exercise(ME),high load exercise(HE),and super high load exercise(SHE)groups.The training groups received 8-week treadmill training,6d/w,1time/d.After training,all the groups were sacrificed to obtain heart samples and myocardial mitochondria were extracted by different centrifugation.The activities of NADH-Q reductase、succinate-Q reductase、cytochrome reductase and cytochrome oxidase were evaluated.Results:Compared with the C group,the NADH-Q reductase activity of ME and SHE groups exhibited increased(P<0.01),while HE group was significantly decreased(P<0.05);the succinate-Q reductase activity of ME group exhibited increased(P<0.05)while HE group exhibited decreased(P<0.01);the cytochrome reductase activity of ME and SHE groups exhibited increased(P<0.01);the cytochrome oxidase activity of ME、HE and SHE groups exhibited increased(P<0.05,P<0.01).Conclusion:Different load exercise has an effect on the function of mitochondrial electron transport chain of myocardial mitochondria.In the respect of improving its function,medium load exercise is the best,followed by super high load exercise.In terms of improving its function,moderate load sports training has the best effect,followed by extremely high load sports training.
