摘要:目的通过建立银屑病大鼠模型,不同剂量龙胆泻肝汤灌胃给药,探讨其对银屑病大鼠皮损及血清白介素-17(interleukin-17,IL-17)、血管内皮生长因子(vascular endothelial growth factor, VEGF)的调节作用。方法将48只大鼠按照简单排序随机化原则分6组,除空白组外,各组大鼠背部进行银屑病皮损造模。造模结束后,生理盐水灌胃给予空白组和模型组,消银颗粒灌胃治疗阳性对照组,龙胆泻肝汤高、中、低不同剂量灌胃治疗其余各组。于治疗开始后对各组大鼠进行表皮厚度及PASI评分;治疗结束后,切取大鼠造模皮肤,进行形态学、组织病理学观察;ELISA法检测各组大鼠血清IL-17、VEGF含量。结果治疗结束后,各治疗组大鼠表皮厚度较模型组均变薄,其中高、中剂量组与阳性对照组相比,具有统计学意义上的下降差异(P<0.01);各治疗组大鼠血清中IL-17、VEGF含量较模型组相比均降低,高、中剂量组与阳性对照组相比,具有统计学意义上的下降差异(P<0.01)。结论灌胃给药不同剂量龙胆泻肝汤后,银屑病大鼠模型的皮损得到不同程度改善,可能与调节大鼠血清IL-17、VEGF有关。
Objective To investigate the effect of Longdan Xiegan Decoction(龙胆泻肝汤) on the expressions of IL-17 and VEGF in psoriatic rats. Methods Forty-eight rats were randomly divided into 6 groups according to the principle of simple order and randomization. After modeling, normal saline was given to blank group and model group by gavage. The positive control group was treated with Xiaoyin Granule(消银颗粒) and the other groups were treated with Longdan Xiegan Decoction at different doses. At the beginning of treatment, the skin thickness and PASI score of rats in each group were measured. After treatment, the model skin was cut off for morphological and histopathological observation. The serum levels of IL-17 and VEGF were detected by ELISA. Result After treatment, the epidermal thickness of each treatment group was thinner than that of the model group, and the high and medium dose groups had a statistically significant decrease compared with the positive control group(P<0.01). The levels of IL-17 and VEGF in the serum of the treatment groups were lower than those of the model group, and the high and medium dose groups’ levels were significantly lower than those of the positive control group(P<0.01). Conclusion After intragastric administration of different doses of Longdan Xiegan Decoction, the skin lesions of psoriasis rat model can be improved in varying degrees, which may be related to the regulation of IL-17 and VEGF in rat serum.
