摘要:目的:观察四神丸对脾肾阳虚型溃疡性结肠炎(ulcerative colitis,UC)模型大鼠Th1/Th2免疫平衡的影响。方法:将90只大鼠随机分为空白组、模型组、对照组、四神丸高剂量组、四神丸中剂量组和四神丸低剂量组,每组各15只。除空白组外,其余各组制备脾肾阳虚证溃疡性结肠炎大鼠模型。造模成功后,空白组则以等体积生理盐水灌胃,对照组以柳氮磺胺嘧啶水溶液0. 5 g·kg-1灌胃,四神丸低剂量组以四神丸药液13 g·kg-1灌胃,四神丸中剂量组以四神丸药液26g·kg-1灌胃,四神丸高剂量组以四神丸药液52 g·kg-1灌胃,模型组灌服等体积生理盐水,共21 d。采用酶联免疫法检测大鼠血清白细胞介素-1β(interleukin-1β,IL-1β)和白细胞介素-4(interleukin-4,IL-4)水平,并计算IL-1β/IL-4;取胸腺组织称质量并计算大鼠胸腺指数; HE染色观察大鼠结肠损伤情况,用免疫组织化学法检测结肠黏膜IL-13的表达。结果:与空白组比较,模型组胸腺指数明显较低,差异具有统计学意义(P <0. 05);与模型组比较,四神丸中剂量组、四神丸高剂量组胸腺指数高于对照组,差异具有统计学意义(P <0. 05)。与空白组比较,模型组和对照组大鼠血清IL-1β含量、IL-1β/IL-4比值显著增高,IL-4含量均降低,差异具有统计学意义(P <0. 05)。与模型组比较,四神丸中剂量组、四神丸高剂量组大鼠IL-1β含量、IL-1β/IL-4比值显著降低,其中以四神丸中剂量组降低明显; IL-4含量显著升高,其中以四神丸中剂量组升高明显,差异有统计学意义(P <0. 05)。与空白组比较,模型组大鼠血清IL-13含量降低,差异具有统计学意义(P <0. 05);与模型组比较,四神丸中剂量组、四神丸高剂量组大鼠结肠组织IL-13蛋白表达增高,其中以四神丸高剂量组表达升高显著,差异具有统计学意义(P <0. 05)。结论:四神丸可提高UC大鼠机体免疫功能,抑制炎症反应,其作用机制可能与调节Th1/Th2免疫平衡有关。
Objective: To observe the influence of Four Spirits Pill on Th1/Th2 immune balance in model rats with spleen kidney yang deficiency ulcerative colitis(UC). Methods: Ninety rats were randomly divided into the blank group,the model group,the control group,the high dose group of Four Spirits Pill,the middle dose group of Four Spirits Pill,the low dose group of Four Spirits Pill,with 15 rats in each group. Except the blank group,the other groups were prepared ulcerative colitis " disease-syndrome combination" rat model of spleen-kidney Yang deficiency syndrome. After successful modeling,the blank group was given the same volume of normal saline,for gavage and intraperitoneal injection. The control group was given 0. 5 g·kg^-1 sulfasalazine solution for gavage. The low dose group of Four Spirits Pill was given13 g·kg^-1 solution for gavage. The middle dose group of Four Spirits Pill was given26 g·kg^-1 solution for gavage. The high dose group of Four Spirits Pill was given 52 g·kg^-1 solution for gavage.The model group was fed with equal volume of normal saline,a total of 21 d. This treatment lasted for 21 days. The serum levels of interleukin-1β(IL-1β) and interleukin-4(IL-4) were measured by enzyme-linked immunosorbent assay(ELISA),and the levels of IL-1 beta/IL-4 were calculated. The thymus tissue was weighed and the thymus index was calculated. HE staining was used to observe the colonic injury in rats,and immunohistochemical method was used to detect the expression of IL-13 in colonic mucosa.Results: Compared with the blank group,the thymus index of the model group was significantly lower(P〈0. 05). Compared with the model group,the thymus indexes of the high dose group of Four Spirits Pill,the middle dose group of Four Spirits Pill,and the difference was statistically significant(P〈0. 05). Compared with the blank group,the serum levels of IL-1βand the ratio of IL-1βto IL-4 in the model group and the control group were significantly increased,and the content of IL-4 was decreased(P〈0. 05).Compared with the model group,the content of IL-1βand the ratio of IL-1βto IL-4 in the middle-dose group and the high-dose group of Four Spirits Pill decreased significantly,especially in the middle-dose group,and the content of IL-4 increased significantly,especially in the middle-dose group(P〈0. 05). Compared with the blank group,the content of IL-13 in the serum of the model group decreased significantly,and the difference was statistically significant(P〈0. 05). Compared with the model group,the expression of IL-13 in the colon tissue of the middle dose group and the high dose group increased significantly,and the difference was statistically significant(P〈0. 05). Conclusion: Four Spirits Pill can improve the immune function and inhibit inflammatory reaction of UC rats,and its mechanism may be related to regulating Th1/Th2 immune balance.
